基于主成分分析法优选半夏、天南星及白附子水丸制剂工艺 被引量:5

Optimization of Preparation Technology of Banxia(Pinelliae Rhizoma),Tiannanxing(Arisaematis Rhizoma)and Baifuzi(Typhonii Rhizoma)Water Pills Based on Principal Component Analysis

摘要 目的利用主成分分析法优选半夏、天南星、白附子生品及炮制品水丸的最佳制剂工艺。方法首先利用均匀设计法设计并制备多批次药材水丸。紫外-可见分光光度法及高效液相色谱法测定水丸中的成分含量(总淀粉、总多糖、总皂苷、尿嘧啶、次黄嘌呤、黄嘌呤、尿苷、鸟苷、夏佛塔苷)。之后利用主成分分析法计算不同样品的综合评价指标值,绘制等高线图并优选出不同水丸的最佳工艺。结果炮制和制剂过程对药品的药性药效有影响。对于3味药材的丸剂来说,半夏生品及最优炮制品水丸的最佳制备工艺为泛丸时间0.5 h、干燥时间8 h、干燥温度80℃,市售炮制品水丸的最佳制备工艺为泛丸时间0.5 h、干燥时间12 h、干燥温度80℃,天南星水丸的最佳制备工艺为泛丸时间0.5 h、干燥时间8 h、干燥温度60℃,白附子水丸的最佳制备工艺为泛丸时间1 h、干燥时间12 h、干燥温度60℃。结论利用主成分分析法,可以聚合多元信息,综合评价,为中药水丸质量判断及工艺优选提供了新的方法。 Objective Principal component analysis(PCA)method was used to optimize the best preparation technology of Banxia(Pinelliae Rhizoma),Tiannanxing(Arisaematis Rhizoma)and Baifuzi(Typhonii Rhizoma)water pills.Method Firstly,the uniform design method was used to design and prepare multiple batches of water pills.Their component contents(total starch,total polysaccharides,total saponins,uracil,hypoxanthine,xanthine,uridine,guanosine,schaffoside)were determined.Secondly,multiple regression models were used to establish regression models between physical parameters and component content,process parameters and component content.Finally,the comprehensive evaluation index values of different pills were calculated by principal component analysis method,contour diagrams were drawn,and the best process was selected.Result It is found that the processing and formulation had an impact on the properties and efficacy of the drugs.The optimal processed product pills of Banxia(Pinelliae Rhizoma)(the raw and the best preparation process)was:pilling time 0.5 h,drying time 8 h,drying temperature 80℃.The best preparation technology of commercially available processed product pills was:pilling time 0.5 h,drying time 12 h,drying temperature 80℃.The best preparation technology of Tiannanxing(Arisaematis Rhizoma)water pills was as follows:pilling time 0.5 h,drying time 8 h,drying temperature 60℃.The best preparation process of Baifuzi(Typhonii Rhizoma)was pilling time 1 h,drying time 12 h and drying temperature 60℃.Conclusion The PCA can be used to aggregate multiple information and synthesize evaluation indexes,which provides a new method for the quality judgmentand process optimizationof water pills.
