摘要
目的新建茯苓显微特征定量方法,明确其与药材活性成分的相关性,为茯苓药材品质评价提供新思路。方法采用容量分析法测定显微特征指数,采用热浸法、紫外分光光度法、HPLC分别测定醇溶性浸出物、水溶性浸出物、多糖以及去氢土莫酸、猪苓酸C、去氢茯苓酸3种三萜类活性成分含量,通过相关分析、主成分分析,评估茯苓显微特征指数在评价药材品质的可行性。结果以称样量30 mg、粉碎粒度120目为茯苓显微特征定量方法最优条件,不同产地间茯苓显微特征指数存在较大差异,范围85.50~145.50千个/mg,与醇溶性浸出物、水溶性浸出物呈显著正相关(P<0.05),与猪苓酸C呈极显著正相关(P<0.01)。经主成分分析,茯苓各指标品质评价贡献大小依次为:显微特征指数﹥浸出物﹥三萜类成分﹥多糖。结论茯苓显微特征指标是药材固有属性,是评价药材品质的重要指标,在科学评价茯苓药材品质过程中具有重要意义。
Objective A quantitative method for the microscopic identification of Fuling(Poria)was developed to clarify the correlation between the microscopic identification and the active components,providing a new idea for quality evaluation of Fuling(Poria).Method The identification index was determined by volumetric analysis,and the contents of alcohol soluble extract,wa⁃ter-soluble extract,polysaccharide and three triterpenoids were determined by hot dipping,UV and HPLC respectively,while the feasibility of microscopic identification in quality evaluation was evaluated by correlation analysis and principal component analy⁃sis.Results The optimal conditions for quantitative analysis of Fuling(Poria)microscopical characteristics as the following:The sample weight was 30 g and crushing mesh was 120.Microscopic characteristic indexes of Fuling(Poria)from different regions were different and ranging from 85.50 to 145.50 hundred per milligram,which were positively correlated with alcohol soluble ex⁃tract and water-soluble extract(P<0.05),and positively correlated with polyporenic acid C(P<0.01).According to the prin⁃cipal component analysis,the intrinsic quality contribution of Fuling(Poria)was as follows:microscopic characteristic indexes>extracts>triterpenoids>polysaccharides.Conclusion The microscopic characteristic index was the inherent attribute of Fuling(Poria)and an important index to evaluate the quality of medicinal materials,which was of great significance in scientific evalua⁃tion of quality of Fuling(Poria).
