摘要
目的半夏白术天麻汤在大鼠心脏的代谢产物研究。方法采取大鼠灌胃给药,建立了半夏白术天麻汤代谢产物的超高效液相色谱-电喷雾-四极杆-飞行时间/质谱(ultra-high-performance liquid chromatography coupled with electrospray ionization quadrupole time-of-flight mass spectrometry,UHPLC-ESI-Q-TOF/MS)分析方法。采用安捷伦SB-C18色谱柱(3 mm×150 mm,1.8μm)。流动相由0.1%甲酸水溶液(A)和乙腈(B)组成,流速为0.3 mL/min。采用0~15 min,15%→90%B的梯度洗脱程序用于分离,质谱采用正、负离子模式检测心脏组织样品。根据分子量以及质谱的碎片离子数据,结合体内代谢反应规律,对复方中各味药中主要成分进行指认和分析研究。结果半夏白术天麻汤各味药中主成分及其代谢产物共鉴定了38个,其中正离子模式下有32个,负离子模式下有6个。大鼠灌胃给药后,半夏白术天麻汤在大鼠心脏中依次发生I相和II相代谢反应,主要的代谢方式为氧化、还原、水解、谷胱甘肽化、亚硫酸化、葡萄糖醛酸化、甲基化和乙酰化。结论首次研究半夏白术天麻汤在大鼠心脏的代谢产物及代谢途径,为半夏白术天麻汤治疗心血管疾病的药理作用机制及临床应用提供科学依据。
Objective To study the metabolites of Banxia Baizhu Tianma Decoction(半夏白术天麻汤)in the heart of rats.Methods The metabolites of Banxia Baizhu Tianma Decoction were established by ultra-high-performance liquid chromatogra⁃phy coupled with electrospray ionization quadrupole time-of-flight mass spectrometry(UHPLC-ESI-Q-TOF/MS)after in⁃tragastric administration.Agilent Zorbax SB-C18(3 mm×150 mm,1.8μm).The mobile phase consists of 0.1%formic acid aqueous solution(a)and acetonitrile(b),and the flow rate was 0.3 mL/min.The gradient elution procedure of 0~15 min and 15%→90%B was used for separation.The positive and negative ion modes were used for mass spectrometry.According to the fragment ion data of molecular weight and mass spectrometry,combined with the law of metabolic reaction in vivo,the main com⁃ponents of each medicine in the compound were identified and analyzed.Results A total of 38 principal components and their me⁃tabolites in Banxia Baizhu Tianma Decoction were identified,including 32 in positive ion mode and 6 in negative ion mode.After intragastrical administration of Banxia Baizhu Tianma Decoction,phase I and phase II metabolic reactions occurred successively in the heart of rats.The main metabolic modes included oxidation,reduction,hydrolysis,glutathione,sulfitation,glucuronidation,methylation and acetylation.Conclusion This paper studies the metabolites and metabolic pathway of Banxia Baizhu Tianma De⁃coction in the heart of rats for the first time,so as to provide a scientific basis for the pharmacological mechanism and clinical ap⁃plication of Banxia Baizhu Tianma Decoction in the treatment of cardiovascular diseases.
