玉米赤霉烯酮对大鼠原代睾丸支持细胞脂质合成的影响

摘要 为揭示玉米赤霉烯酮(zearalenone,ZEA)的雄性生殖毒性及其机理,本试验研究了ZEA对大鼠睾丸支持细胞(sertoli cells,SCs)脂质合成的影响。选用大鼠原代SCs作为材料,设置对照组(无ZEA组)、染毒组(不同浓度ZEA处理组),处理24 h后,CCK-8法检测细胞活性,甘油三酯(TG)试剂盒检测TG含量,尼罗红染色检测细胞内脂滴含量,蛋白免疫印迹检测脂肪酸合成蛋白的表达,ATP试剂盒检测细胞内ATP含量。结果显示,染毒组与对照组相比,ZEA浓度为20,30μmol/L时,大鼠SCs活性呈极显著下降趋势;10,20μmol/L ZEA可促使SCs内TG含量的极显著上升(P<0.01),且呈剂量依赖性关系;20μmol/L ZEA可促使SCs内脂滴含量的极显著性上升(P<0.01);各ZEA染毒组的脂质合成相关蛋白PPARγ和SREBP1的表达呈现极显著性上升(P<0.01),FAS呈现上升趋势但无显著性(P>0.05);10,20μmol/L ZEA组ATP呈现极显著性下降(P<0.01)。结果表明,ZEA对大鼠SCs具有一定的毒性,可干扰SCs的脂质合成和ATP生成,从而影响雄性生殖细胞的能量供应。 Lipid synthesis is an important part of the energy supply of rat Sertoli cells.In order to explore the reproductive toxicity of zearalenone,the effects of ZEA on lipid synthesis of rat primary Sertoli cells(SCs)were investigated.Testicular Sertoli cells were used as the material in this experiment.The control group(without ZEA group)and a test group(treated with different concentrations of ZEA)were set up.After 24 h of treatment,the CCK-8 method was used to detect the cell viability,the triglyceride kit was used to detect the triglyceride content,Nile Red staining was used to detect the intracellular lipid content,Western blot was used to detect fatty acid synthesis protein expression,ATP kit was used to detect the intracellular ATP content.The results showed that compared with the control group,when the ZEA concentration was 20 and 30μmol/L,cell viability was extremely significantly reduced.ZEA at 10 and 20μmol/L promoted a very significant increase in intracellular TG content(P<0.01),which showed dose-dependent relationship.20μmol/L ZEA promoted a very significant increase in the lipid droplet content in testicular Sertoli cells(P<0.01).The expression of lipid synthesis-related proteins PPARγand SREBP1 in the ZEA-exposed group showed a significant rising trend.FAS showed an upward trend but not significant.The ATP of the ZEA group of 10 and 20μmol/L showed a very significant decrease(P<0.01).It is concluded that zearalenone is toxic to rat Sertoli cells.Within a certain concentration range,it can interfere with the lipid synthesis of SCs and interfere with the ATP production of SCs.
