摘要
以海藻酸钠、壳聚糖、生物活性玻璃为主要原料制备海洋多糖复合水凝胶,分别以成胶时间、含水量、溶胀率、力学性能为指标进行性能测试,并利用扫描电镜分析复合水凝胶的微观表面形貌。实验结果表明,当海藻酸钠与壳聚糖体积比为17∶3时,复合水凝胶成胶时间最短,约为60 s。生物活性玻璃的添加对复合水凝胶成胶时间影响不大,但有助于使其网状结构更加紧密、分散性更加均匀,其力学性能远高于未添加生物活性玻璃的复合水凝胶。同时,复合水凝胶的含水量、溶胀率随海藻酸钠与壳聚糖体积比的减小而减小,随生物活性玻璃添加量的增加而降低。据此筛选出复合水凝胶制备最适条件为:海藻酸钠与壳聚糖体积比为17∶3,生物活性玻璃质量浓度为0.5 g/mL。将南极磷虾抗菌肽负载于海洋多糖复合水凝胶上,并探究其对金黄色葡萄球菌和变异链球菌的抑制作用,得到了具有良好抑制细菌活性的海洋多糖/抗菌肽复合抗菌水凝胶产物。
The composite hydrogel of marine polysaccharide was prepared by using sodium alginate, chitosan, and bioactive glass as the main raw materials. The gelation time, water content, swelling rate, and mechanical properties were used as the performance test indexes. And the microscopic surface structure of composite hydrogel was analyzed by scanning electron microscopy. The results indicated that the shortest gelation time of composite hydrogel was about 60 s when the volume ratio of sodium alginate and chitosan was 17∶3. The addition of bioactive glass showed little effect on the gelation time of composite hydrogel. But it contributed to forming composite hydrogel with the tighter network structure and the more uniform dispersion. The mechanical property was much better than that of composite hydrogel without bioactive glass. At the same time, the water content and swelling rate of composite hydrogel decreased with the reduction of the volume ratio of sodium alginate and chitosan, and decreased with the increase of bioactive glass. Based on the results, the optimal conditions for the preparation of composite hydrogel were as follows: V(sodium alginate)∶V(chitosan) was 17∶3, and the concentration of bioactive glass was 0.5 g/mL in this experiment. The antibacterial peptide of the Antarctic krill was loaded on the marine polysaccharide composite hydrogel. The antibacterial effect was investigated against Staphylococcus aureus and Streptococcus mutans. The marine polysaccharide/antibacterial peptide composite hydrogel was obtained with the better antimicrobial activity.
