褐藻素对乳腺癌MDA-MB-231细胞增殖和凋亡的影响

摘要 目的:探究褐藻素对人乳腺癌MDA-MB-231细胞的增殖和凋亡的影响。方法:不同浓度褐藻素处理MDA-MB-231细胞后,采用CCK-8法测定细胞抑制率、Live/DeadTM细胞成像试剂盒进行活死细胞荧光分析,TUNEL法、Annexin V/PI染色法检测细胞凋亡,Western blotting方法检测Bcl-2、Bax、JAK2、p-JAK2、STAT3、p-STAT3蛋白的表达,采用试剂盒检测Caspase-9和Caspase-3/7活性。结果:褐藻素处理MDA-MB-231细胞24 h时,16μmol·L^(-1)褐藻素明显抑制细胞增殖,IC_(50)为68.79μmol·L^(-1);处理48 h时,8μmol·L^(-1)褐藻素明显抑制细胞增殖,IC_(50)为38.08μmol·L^(-1);褐藻素对乳腺癌细胞的细胞活力和增殖的抑制作用表现出一定的时间和剂量依赖性(P<0.05)。与阴性对照组相比,16,32,64μmol·L^(-1),褐藻素可上调Bax蛋白表达,下调Bcl-2蛋白表达,提高Caspase-9和Caspase-3/7活性,诱导细胞凋亡,且呈现一定的剂量依赖性(P<0.05),还可以抑制JAK2、STAT3蛋白的磷酸化激活(P<0.05)。结论:褐藻素抑制乳腺癌MDA-MB-231细胞增殖,诱导其凋亡,机制可能和抑制JAK2、STAT3蛋白磷酸化激活有关。 Objective: To investigate the effects of fucoxanthin on the proliferation and apoptosis of human breast cancer MDA-MB-231 cells. Methods: MDA-MB-231 cells were treated with different concentrations of fucoxanthin. The cell inhibition rate was determined by CCK-8 method, the fluorescence analysis of living and dead cells was performed by Live/Deadtmcell imaging kit, the apoptosis was detected by TUNEL method and Annexin V/PI staining method, the expression of Bcl-2, Bax, JAK2, p-JAK2, STAT3, and p-STAT3 and proteins was detected by Western blotting method, and the activities of Caspase-9 and Caspase-3/7 were detected by corresponding kit.Results: MDA-MB-231 cells were treated with fucoxanthin for 24 h, and 16 μmol·L^(-1)fucoxanthin could significantly inhibit the cell proliferation with IC_(50)value of 68.79 μmol·L^(-1);8 μmol·L^(-1)fucoxanthin could significantly inhibit the cell proliferation after 48 h treatment with IC_(50)value of 38.08 μmol·L^(-1). The fucoxanthin-treated MDA-MB-231 cells showed dose-and time-dependent decrease in the cell viability and proliferation. Fucoxanthin(16, 32, 64 μmol·L^(-1)) could up-regulate the expression of Bax protein, down-regulate the expression of Bcl-2 protein, increase the activities of Caspase-9 and Caspase-3/7, and induce the apoptosis in dose-and time-dependent manners(P<0.05);fucoxanthin could also inhibit the phosphorylation activation of JAK2 and STAT3 proteins(P<0.05). Conclusion: Fucoxanthin can inhibit the proliferation and induce the apoptosis of MDA-MB-231 cells, which may be related to the inhibition of the phosphorylation activation of JAK2 and STAT3 proteins.
