摘要
为了研究慢性冷暴露对小鼠肾脏的危害,将试验小鼠随机设置为对照组和冷暴露组,室温对照组小鼠在(24±2)℃的环境中饲养,冷暴露组小鼠在早上8点至晚上8点期间随机于4℃的环境中刺激3 h,共刺激3周,用全自动化分析仪检测小鼠肾功能相关血液生化指标。结果表明冷暴露会影响肾脏的功能。进一步应用q-PCR和蛋白质免疫印迹技术,测定小鼠肾脏炎症相关因子mRNA的表达、内质网应激(ERS)和自噬相关蛋白质的表达水平。研究结果显示与对照组比较,冷暴露组的小鼠肾脏炎症因子mRNA的表达、ERS和自噬相关蛋白质表达都显著升高。结果表明,冷暴露会促进小鼠肾脏产生炎症,引发ERS,诱发自噬。
In order to study the harm of chronic cold exposure to the kidneys of mice,the experimental mice were randomly set as the control group and the cold exposure group.The mice in the room temperature control group were raised in an environment of(24±2)℃,and the mice in the cold exposure group were randomly stimulated in an environment of 4℃for 3 h from 8 am to 8 pm,and co-stimulated for 3 weeks.A fully automated analyzer is used to detect blood biochemical indicators related to renal function.The results show that cold exposure can affect the function of the kidneys.Furthermore,q-PCR and Western blot techniques were used to determine the expression levels of mouse kidney inflammation-related factors mRNA,endoplasmic reticulum stress(ERS)and autophagy-related proteins.The results of the study showed that compared with the control group,the expression of renal inflammatory factors mRNA,ERS and autophagy-related protein expression of mice in the cold exposure group were significantly increased.The results showed that cold exposure can promote inflammation in the kidneys of mice,trigger ERS,induce autophagy,and cause damage to the kidneys.
