摘要
本研究以5个萱草品种为试验材料,分析其形态差异并明确不同萱草品种花粉生活力和柱头可授性,为有效提高萱草杂交育种提供基础。研究结果表明,‘H5’花朵直径最大,即将开放时的花蕾也最长,‘H2’花朵直径和即将开放时的花蕾最小;‘H4’花葶最高,‘H3’(‘金娃娃’)最矮;‘H2’株高最高,‘H3’最矮;‘H1’叶片长宽比最大,‘H3’最小。染色结果表明,醋酸洋红、KI-I2和TTC染色液均能使萱草花粉着色,且不同品种间存在显著性的差异。花粉离体培养结果表明‘H1’花粉在36种组合培养基中均不能正常萌发。柱头可授性结果表明,‘H1’和‘H5’柱头可授性较高,‘H2’,‘H3’和‘H4’依次降低。综合以上研究发现‘H1’和‘H5’是萱草杂交育种中很好的亲本材料。本研究通过花粉生活力和柱头可授性的鉴定可快速筛选杂交育种中的优良父母本,为萱草新品种的培育提供便利。
In this study, five Hemerocallis fulva cultivars were used as materials to analyze the morphological differences, pollen viability and stigma receptivity, so as to lay the foundation for improving the hybrid breeding efficiency in Hemerocallis fulva. The results showed that the flower diameter and flower bud before opening of ’H5’ is the largest and longest, while ’H2’ is the smallest and shortest. The height of scape of ’H4’ is the tallest, but ’H3’(’Gold baby’) is the shortest. The plant height of ’H2’ is the highest, however ’H3’ is the shortest. The ratio blade length to width of ’H1’ is the maximal, nevertheless ’H3’ is the minimum. The staining results showed that the pollens of five Hemerocallis fulva cultivars could be color by the dye solution of aceto-carmine, KI-I2and TTC, moreover, the difference was between various cultivars. The result of optimal medium for pollens germination showed that the pollens of ’H1’ could not normally germinate in all thirty-six combination media. The result of stigma receptivity show that ’H1’ and ’H5’ was higher, while ’H2’, ’H3’ and ’H4’ was gradually decreased. In general, ’H1’ and’H5’ are the excellent parent materials for crossbreeding in Hemerocallis fulva. It will quickly screen the admirable parents for hybrid breeding and provides convenience for the cultivation of new varieties in Hemerocallis fulva through the identification of pollen viability and stigma receptivity in this study.
