摘要
目的 探讨宫颈癌(CC)组织中含Jumonji结构域的蛋白2B(JMJD2B)、造血相关PBX相互作用蛋白(HPIP)的表达与患者预后的关系。方法 选取2014年6月至2016年6月唐山市妇幼保健院收治的84例CC患者作为研究对象。采用免疫组织化学法检测CC组织中JMJD2B、HPIP蛋白表达情况,分析JMJD2B、HPIP蛋白表达与临床病理特征的关系。采用定量聚合酶链反应(PCR)法检测CC组织中JMJD2B、HPIP mRNA表达水平,分析各相关因素与CC患者5年总存活率(OS)的关系。结果 84例CC组织样本中,JMJD2B、HPIP蛋白阳性表达率分别为60.71%和54.76%;不同病理类型、国际妇产科联盟(FIGO)分期、有无淋巴节转移及不同分化程度CC组织中JMJD2B、HPIP蛋白阳性表达率均存在显著差异(P<0.05)。CC组织中JMJD2B、HPIP mRNA表达水平均显著高于CC癌旁组织(P<0.05);FIGO分期高、有淋巴节转移、分化程度低及JMJD2B、HPIP阳性表达均是CC患者5年OS的独立危险因素(P<0.05)。结论 CC组织中JMJD2B、HPIP表达均显著上调,且均是CC患者的独立预后指标。
Objective To investigate the relationship between the expression of Jumonji domain containing protein 2B(JMJD2B) and hematopoietic associated PBX interaction protein(HPIP) in cervical cancer(CC) and the prognosis of patients. Methods 84 CC patients admitted to Tangshan Maternal and Child Health Hospital from June 2014 to June 2016 were selected as the study subjects. Immunohistochemistry was used to detect the expression of JMJD2B and HPIP proteins in CC tissues, and the relationship between the expression of JMJD2B and HPIP proteins and clinicopathological characteristics was analyzed. Quantitative polymerase chain reaction(PCR) was used to detect the expression level of JMJD2B and HPIP mRNA in CC tissues and analyze the relationship between each related factor and the 5-year overall survival rate(OS) of CC patients. Results In 84 CC tissue samples, the positive expression rates of JMJD2B and HPIP protein were 60.71% and 54.76% respectively;the positive expression rates of JMJD2B and HPIP protein in CC tissues of different pathological types, international federation of gynecology and obstetrics(FIGO) staging, lymph node metastasis and different differentiation levels were significantly different(P<0.05). The expression levels of JMJD2B and HPIP mRNA in CC tissues were significantly higher than those in CC adjacent tissues(P<0.05);high FIGO stage, lymph node metastasis, low degree of differentiation and positive expression of JMJD2B and HPIP were independent risk factors of OS in CC patients for 5 years(P<0.05). Conclusions The expression of JMJD2B and HPIP in CC tissues were significantly up-regulated, and they were independent prognostic indicators of CC patients.
