摘要
2019年7月山西首次发现草地贪夜蛾入侵,当地农业生产受到严重影响。采用细胞色素C氧化酶亚基Ⅰ(COⅠ)和磷酸甘油醛异构酶(Tpi)的两个分子标记对山西草地贪夜蛾样本进行遗传特征分析。基于母系遗传特征COⅠ基因片段的序列分析表明,样本中水稻型所占比例为97.6%,玉米型所占比例为2.4%;基于父系遗传特征Tpi基因片段的序列分析则表明所有样本均为玉米型,并在174、175位点存在3种碱基特征:87.80%的样本为玉米型碱基特征(AT),9.76%为水稻型碱基特征(GA),2.43%为杂合型特征(AA)。由此可见,入侵山西的草地贪夜蛾主要是母系水稻型和父系玉米型的杂合型。研究结果可为研究草地贪夜蛾迁飞过程中的种群遗传特征变化提供理论依据,为农业部门监测及制定防控策略提供参考。
The fall armyworm, Spodoptera frugiper da, was detected for the first time in Shanxi in July 2019, and threatened local agricultural production. Two molecular markers of cytochrome c oxidase subunit(CO Ⅰ) and triose-phosphate isomerase(Tpi) were used to analyze the genetic characteristics of the population of S. frugiperda in Shanxi.The CO Ⅰ gene analysis showed that 97.6% of samples were the rice-strain and 2.6% were the corn-strain. Sequence analysis based on Tpi gene fragments showed that all the samples were the corn-strain, and there were three different types at polymorphic sites 174 and 175, 87.80% of the samples were corn-strain(AT), 9.76% were rice-strain(GA), and 2.43% were hybrid type(AA). The results indicated that the invasion of S. frugiperda in Shanxi was mainly heterozygous,which provided a theoretical basis for studying the changes in genetic characteristics during migration, and a reference for monitoring and developing control strategies.
