摘要
目的:基于“形寒寒饮则伤肺”理论建立寒饮蕴肺证大鼠病理模型,并从宏观功能学及微观分子学角度进行可靠性评价。方法:应用冷冻方法施加“形寒”、灌胃冰水方法施加“饮冷”及冷水浴施加“劳则气耗”等因素,复制寒饮蕴肺证大鼠病理模型。通过观察大鼠体质量及一般行为学、肺组织病理形态学变化,肺功能分析系统检测肺功能参数,ELISA法检测肺组织环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、肿瘤坏死因子-α(TNF-α)表达,免疫组化法检测水通道蛋白5(AQP5)、黏蛋白5AC(MUC5AC)的表达,以对模型进行可靠性评价。结果:与对照组比较,模型组大鼠在宏观功能表现上,出现明显的寒饮蕴肺证症状及体征,体质量差值显著下降(P<0.01),肺功能各指标显著改变(P<0.01);在微观分子表达上,cAMP/cGMP比值显著减低(P<0.01),TNF-α表达显著升高(P<0.01),AQP5表达显著降低(P<0.01)。结论:该模型的致病因素、病理改变、实验室指标及临床特征方面均与临床典型的寒饮蕴肺证相似,通过该方法建立的寒饮蕴肺证大鼠病理模型是一种贴近自然发生疾病的病理模型,具有可行性与可靠性。
Objective:To establish the pathological model of the rat with syndrome of accumulation of cold fluid in the lung based on the theory of‘cold body and cold excessive fluid can lead to lung injury’,and to evaluate the reliability of the model from the perspectives of macro-function and molecular analysis.Methods:The pathological model of the syndrome of accumulation of cold fluid in the lung was established by applying the stimulation factors such as‘cold body’by freezing,‘cold excessive fluid’by intragastric administration of ice water,and‘over-strain consuming essence’by cold bath.The weight,general behavior and the pathological changes of lung tissue of rats were observed.The lung function was detected by lung function analysis system.The expressions of cyclic adenosine monophosphate(cAMP),cyclic guanosine monophosphate(cGMP)and tumor necrosis factor-α(TNF-α)in lung tissue were detected by enzyme-linked immunosorbent assay(ELISA).The expressions of aquaporin(AQP)5 and mucin(MUC)5AC were detected by immunohistochemistry,so as to evaluate the reliability of the model.Results:Compared with the control group,the rats in the model group showed obvious symptoms and signs of syndrome of accumulation of cold fluid in the lung in the macro-functional performance,and their weight decreased significantly(P<0.01),and their lung function decreased(P<0.01).In terms of microscopic molecular expression,the ratio of cAMP to cGMP was significantly decreased(P<0.01),the expressions of TNF-αwas increased(P<0.01),and the expression of AQP5 was decreased(P<0.01).Conclusion:The pathogenic factors,pathological changes,laboratory indicators and clinical features of the model are similar to those of the typical clinical syndrome of accumulation of cold fluid in the lung.The pathological model of the rat with syndrome of accumulation of cold fluid in the lung established by this method is a pathological model close to the characteristics of human spontaneous diseases,which has feasibility and reliability.
