摘要
建立高效液相色谱法(HPLC)测定阿扎胞苷中残留溶剂二甲基亚砜(DMSO)含量。采用高效液相色谱法,色谱柱为Agilent ZORBAX C8(4.6×250mm,5um);流动相为水-乙腈95∶5(V∶V),等度洗脱;检测波长为214nm;流速1.0mL/min。二甲基亚砜与相邻杂质峰分离度良好,在5.060~48.576μg/mL浓度范围内,线性关系良好(r=0.9995),定量限为5.06μg/mL,检测限为0.12μg/mL,平均回收率为100.5%。
A high performance liquid chromatography(HPLC) method was established for the determination of residual solvent dimethyl sulfoxide(DMSO) in azacytoside. HPLC was performed on Agilent ZORBAX C8(4.6×250 mm, 5 um) column. The mobile phase was water-acetonitrile 95:5(V:V), equal degree elution;The detection wavelength was 214 nm;Flow rate 1.0mL0nmR-1. The peak separation between DSO and adjacent impurities was good, and the linear relationship was good(r = 0.999 5)in the concentration range of 5.060 μg/ml to 48.576 μg/ml. The limit of quantitation was 5.06 μg/ml, the detection limit was 0.12 μg/ml, and the average recovery was 100.5%.
