PKM2基因在乳腺癌中的表达及临床病理特征分析 被引量:2

摘要 目的 检测M2型丙酮酸激酶(PKM2)分子在乳腺癌组织中的表达,并探讨PKM2与乳腺癌病情进展与预后的关系。方法 选取2018年10月至2020年10月在山东第一医科大学第二附属医院行手术治疗的乳腺癌患者63例,采用Real-time PCR方法检测PKM2 mRNA在乳腺癌组织、癌旁组织的表达,并分析各亚组间PKM2表达的差异及其与乳腺癌临床病理参数间的关系。结果 在乳腺癌组织中,高肿瘤增殖指数(Ki-67)组表达高于低Ki-67组,淋巴结阳性组表达高于淋巴结阴性组,但差异均无统计学意义(P> 0.05);PKM2 mRNA在乳腺癌组织中的表达量显著高于癌旁组织,差异有统计学意义(P <0.05)。低Ki-67组、高Ki-67组PKM2 mRNA在乳腺癌组织中的表达量均高于乳腺癌旁组织,差异均有统计学意义(P <0.05);淋巴结阴性组、淋巴结阳性组PKM2 mRNA在乳腺癌组织中的表达量均高于乳腺癌旁组织,差异均有统计学意义(P <0.05)。结论 PKM2 mRNA在乳腺癌组织中高表达,而在Ki-67、淋巴结转移的乳腺癌组织中表达各不相同,提示PKM2基因很可能是乳腺癌靶向治疗新的潜在靶点。 Objective To detect the expression of pyruvate kinase M2(PKM2) in breast cancer tissue and explore the relationship between PKM2 and the progression and prognosis of breast cancer. Methods A total of 63 patients with breast cancer who underwent surgical treatment in the Second Affiliated Hospital of Shandong First Medical University from October 2018 to October 2020 were selected. The expression of PKM2 mRNA in breast cancer tissues and cancer-adjacent tissues was detected by Real-time PCR, and the difference of PKM2 expression among various subgroups and its relationship with clinicopathological parameters of breast cancer were analyzed. Results The expression of PKM2 mRNA in breast cancer tissues of the high tumor proliferation index(Ki-67) group was higher than that of the low Ki-67 group, and that of the positive lymph node group was higher than that of the negative lymph node group, with no statistically significant differences(P > 0.05). The expression level of PKM2 mRNA in breast cancer tissues was significantly higher than that in the cancer-adjacent tissues, with statistically significant difference(P < 0.05). In the low Ki-67 group and the high Ki-67 group, the expression level of PKM2 mRNA in breast cancer tissues was higher than that in human breast cancer-adjacent tissues, with statistically significant difference(P < 0.05). In the negative lymph node group and the positive lymph node group, the expression level of PKM2 mRNA in breast cancer tissues was higher than that in human breast cancer-adjacent tissues, with statistically significant difference(P < 0.05).Conclusion PKM2 mRNA is highly expressed in breast cancer tissues, while it is differentially expressed in breast cancer tissues with different Ki-67 and lymph node metastasis status, suggesting that PKM2 gene may be a new potential target for targeted therapy of breast cancer.
