摘要
目的利用多种数据库联合分析肺腺癌中FAM83A基因的表达水平及其临床意义。方法通过多种数据库联合,对肺腺癌组织与正常肺组织中FAM83A基因表达情况进行分析,包括总生存期分析、单因素及多因素回归分析、FAM83A表达水平与肺腺癌临床和病理学特征的相关性分析。分析FAM83A基因与肺腺癌单个细胞功能状态的相关性、FAM83A相互作用的蛋白网络及与临近LncRNA的相关性。结果肺腺癌组织中FAM83A基因水平显著升高(P<0.01);不同年龄、不同分期、有无淋巴转移、有无吸烟史患者(P均<0.01)的肺腺癌组织中FAM83A基因水平显著不同;FAM83A高表达患者较低表达患者而言,总体生存期及无进展生存期显著缩短(P均<0.01)。此外,FAM83A是胰腺癌患者预后的独立危险因素(P<0.001)。FAM83A基因的表达与细胞缺氧呈正相关。与FAM83A相互作用的蛋白有EGFR、RAF1等。FAM83A与其临近的lncRNA FAM83A-AS1存在相关性(r=0.85,P<0.05)。结论FAM83A在肺腺癌组织中表达上调,其基因水平与肺腺癌患者的年龄、吸烟史、恶性程度以及不良预后相关,其作为驱动基因在肺腺癌的发生发展过程中发挥作用,有望成为肺腺癌治疗及预后评估的新靶点。
Objective To analyze the expression level of FAM83A gene in patients with lung adenocarcinoma(LUAD)and its clinical significance based on multidata base.Methods Overall survival analysis,univariate and multivariate regression analysis were applied to analyze the gene expression level of FAM83A gene in lung adenocarcinoma tissues and normal tissues based on the multidata base.The correlation between the expression level of FAM83A with clinical and pathological characteristics of LUAD was further analyzed.The correlation between FAM83A and functional status of single cell of lung adenocarcinoma was analyzed.The adjacent lncRNAs and protein networks interacting with FAM83A was analyzed.Results The FAM83A gene level in lung adenocarcinoma tissues was significantly higher than that in normal lung tissues(P<0.01).The gene levels of FAM83A in lung adenocarcinoma were significantly difference between different ages,lymph node metastasis,smoking history(P<0.01)and stage.The patients with high expression of FAM83A level had shorter overall survival and progression-free survival than those with low expression level.In addition,FAM83A was an independent risk factor for the prognosis of LUAD patients,which was an independent prognostic factor within the range of HR(P<0.01).FAM83A gene was positively correlated with hypoxia.The proteins that interact with FAM83A include EGFR,RAF1,etc.FAM83A was correlated with its adjacent lncRNA FAM83A-AS1,and it was statistically significant(r=0.85,P<0.05).Conclusion FAM83A is highly expressed in cancerous tissues in patients with LUAD,showing a driver gene of LUAD,and those with higher FAM83A gene level is associated with ages,smoking history,stage and poor prognosis.It was expected to become a new targets for treatment and prognosis of LUAD.
