摘要
在室内对苜蓿轮枝菌菌株Ms197和Ms198的最适生长温度、pH值、碳源、氮源、产毒能力及致病力进行测定,同时利用蘸根接种法对37份苜蓿材料的抗病水平进行评价。结果表明,苜蓿轮枝菌菌株Ms197和Ms198在温度5~35℃和pH值4~11范围内均能生长和产孢,具有较强的温度和酸碱度适应性,在温度5~35℃范围内菌落生长速率分别为0.20~4.14 mm/d和0.48~4.12 mm/d,产孢量分别为3.17×10^(5)~4.12×10^(8)和1.02×10^(6)~4.27×10^(8)。在pH值4~11范围内菌落生长速率分别为1.26~3.96 mm/d和1.39~3.97 mm/d,产孢量分别为5.5×10^(5)~4.35×10^(8)和7.67×10^(5)~4.70×10^(8)。最适菌落生长温度和pH值分别为25℃和6~9,最适产孢温度和pH值分别为20℃和9;可利用多种碳源和氮源生长,综合来看,碳、氮源分别为可溶性淀粉和牛肉膏的培养基最适于菌落生长,碳、氮源分别为D-牛乳糖和胰蛋白胨的培养基最适于菌株产孢。菌株Ms197和Ms198的产毒能力和致病力无显著差异。将菌株Ms198接种于37份苜蓿材料,28 d后均表现出不同程度的黄萎病症状,发病率、死亡率和病级分别介于5.33%~100.00%、0.00%~88.20%和1.05~4.88之间;表现高抗、中抗、中感和高感的材料分别为14份、8份、7份和8份,占比分别为37.84%、21.62%、18.92%和21.62%,未发现免疫品种。
In the laboratory,the optimum growth temperature,pH value,carbon source,nitrogen source,toxigenic ability and pathogenicity of Verticillium alfalfae Ms197 and Ms198 were determined,and the disease resistance level of 37 alfalfa varieties was evaluated by root dipping inoculation.The results showed that both strain Ms197 and Ms198 could grow and produce spores in the temperature range of 5~35℃and pH value of 4~11,and had strong adaptability to temperature and pH.In the temperature range of 5~35℃,the colony growth rates of strain Ms197 and Ms198 were 0.20~4.14 mm/d and 0.48~4.12 mm/d,respectively,and the spore production were 3.17×10^(5)~4.12×10^(8)and 1.02×10^(6)~4.27×10^(8),respectively.In the pH value of 4~11,the colony growth rates of strain Ms197 and Ms198 were 1.26~3.96 mm/d and 1.39~3.97 mm/d,respectively,and the spore production were 5.50×10^(5)~4.35×10^(8)and 7.67×10^(5)~4.70×10^(8),respectively.The optimum temperature and pH for colony growth were 25℃and 6~9,respectively,and the optimum temperature and pH for sporulation were 20℃and 9,respectively.A variety of carbon and nitrogen sources can be used to grow the colony,of which the medium with soluble starch and beet extract as carbon and nitrogen sources respectively was the most suitable for colony growth,and the medium with D-lactose and tryptone as carbon and nitrogen sources respectively was the most suitable for spore production.There was no significant difference in toxigenicity and pathogenicity between strains Ms197 and Ms198.After inoculating 37 alfalfa varieties with strain Ms198 for 28 days,all of them showed different degrees of Verticillium wilt symptoms.The incidence rate,mortality and disease grade were between 5.33%to 100.00%,0.00%to 88.20%and 1.05 to 4.88,respectively.There were 14,8,7,and 8 varieties showed high resistance,medium resistance,medium susceptibility and high susceptibility,respectively,accounting for 37.84%,21.62%,18.92%and 21.62%,respectively,but no immune varieties were found.
