紫花苜蓿根腐病病原菌分离鉴定与致病性 被引量:12

摘要 为明确苜蓿根腐病病原菌种类及其致病性,分离纯化黑龙江八一农垦大学安达试验田有根腐病表现的苜蓿根部样品上的真菌,采用形态学观察及rDNA-ITS、theβ-tubulin gene(TUB)序列分析鉴定其种类,并将各种真菌回接于龙牧801苜蓿植株,测定根长比、苗长比、发病率和病情指数,评价其致病性。结果表明,从患病苜蓿根部共分离出7种真菌,经鉴定分别为腐皮镰刀菌(Fusarium solani)、轮枝镰刀菌(F.verticillioides)、尖孢镰刀菌(F.oxysporum)、层出镰刀菌(F.proliferatum)、三线镰刀菌(F.tricinctum)、芬芳镰刀菌(F.redolens)和粉红单端孢(Trichothecium roseum),分离率最高的为腐皮镰刀菌(29.73%),其他真菌分离率在0.68%~20.27%之间。苜蓿接种不同致病菌后,根长比、苗长比、发病率和病情指数均存在显著差异(P<0.05)。采用隶属函数法对不同病原菌进行致病性综合评价,结果表明不同病原菌致病性为:尖孢镰刀菌>芬芳镰刀菌>三线镰刀菌>腐皮镰刀菌>层出镰刀菌>轮枝镰刀菌>粉红单端孢。 In order to identify the species and pathogenicity of alfalfa root rot pathogens,fungi were isolated and purified from the alfalfa root samples with root rot in the Anda experimental field of Heilongjiang Bayi Agricultural University.The species were identified by morphological observation and sequence analysis of rDNA-ITS and theβ-tubulin gene(TUB),and fungi were inoculated back into Longmu 801 alfalfa to determine root length ratio,seedling length ratio,incidence rate and disease index,and evaluate its pathogenicity.The results showed that a total of 7 fungi were isolated from the root of diseased alfalfa.They were identified as Fusarium solani,F.verticillioides,F.oxysporum,F.proliferatum,F.tricinctum,F.redolens and Trichothecium roseum.F.solani had the highest isolation rate(29.73%),and the isolation rates of other fungi ranged from 0.68%tO_(2)0.27%.There were significant differences in root length ratio,seedling length ratio,incidence rate and disease index in alfalfa inoculated with different pathogenic fungi(P<0.05).The membership function was used to evaluate the pathogenicity of different pathogens,the results showed the pathogenicity of different pathogens was as follows:F.oxysporum>F.redolens>F.tricinctum>F.solani>F.proliferatum>F.verticillioides>T.roseum.
