犏牛INHBB基因的克隆及在附睾不同区段的表达研究

摘要 抑制素β_(B)亚基基因(Inhibin beta B,INHBB)对哺乳动物的生殖调控至关重要。为探究INHBB基因与犏牛不育的可能联系,对犏牛附睾INHBB基因进行克隆、生物信息学分析,并采用实时荧光定量PCR分析INHBB基因在犏牛附睾不同区段的表达情况。结果表明,犏牛INHBB基因的CDS序列长度1227 bp,共编码408个氨基酸;INHBB蛋白是亲水蛋白且没有跨膜结构,分子式C_(1990)H_(3128)N_(580)O_(569)S_(22),相对分子量和等电点分别为44.99 ku和8.72;犏牛INHBB基因核苷酸序列与绵羊、山羊等物种有较高的种间同源性,分别为97.4%和97.3%;INHBB基因在附睾头部表达最高(P<0.05),体部次之,尾部表达量相对较低。推测INHBB基因对精子成熟过程有一定的作用。 The Inhibin subunit beta B gene(INHBB)is essential for the regulation of mammalian reproduction.To study the possible link between INHBB gene and sterility of cattle-yak,in this study,the INHBB gene was cloned,bioinformatically analyzed and the expression of the INHBB gene in different segments of the epididymis was analyzed by real-time fluorescence quantitative PCR.The results showed that the CDS sequence of the INHBB gene was 1227 bp in length,encoding 408 amino acids;INHBB protein was hydrophilic and had no transmembrane structure,it's molecular formula was C_(1990)H_(3128)N_(580)O_(569)S_(22),and it's molecular weight and isoelectric point were 44.99 ku and 8.72;the nucleotide sequence of the INHBB gene of cattle-yak had high interspecific homology with sheep and goats,which were 97.4%and 97.3%respectively;the expression of INHBB gene was highest in epididymis caput(P<0.05),but lower in corpus and cauda.It is speculated that the INHBB gene has a certain role in the sperm maturation process.
