摘要
目的 建立液质联用法测定胶原蛋白海绵中L-羟脯氨酸的方法。方法 采用6 mol·L^(-1)盐酸-0.5%苯酚酸解胶原蛋白海绵,以乙腈-0.1%甲酸(5∶95)为流动相,质谱测定特征氨基酸L-羟脯氨酸,以茶氨酸为内标定量分析。结果 方法检出限为0.02 ng,线性范围为2.04~30.6μg·m L^(-1),方法的重复性RSD为2.4%,准确度为98.3%(RSD=2.3%)。结论 本方法准确可靠,可作为胶原蛋白类产品的含量测定方法。
OBJECTIVE To develop a method for determination of L-hydroxyproline in Collagen sponge by liquid mass spectrometry.METHODS Collagen sponge was degraded by 6 mol·L^(-1)hydrochloric acid-0.5%phenol,and acetonitrile-0.1%formic acid (5∶95) was used as mobile phase.L-hydroxyproline was determined by mass spectrometry with the anine as internal standard quantitative analysis.RESULTS The detection limit was0.02ng,the linear range was 2.04-30.6μg·m L^(-1),the repeatability RSD was 2.4%,and the accuracy was98.3%(RSD=2.3%).CONCLUSION This method is accurate and reliable,and can be used as the content determination method for collagen products.
