miR-181a-5p及MEST在高级别浆液性卵巢癌组织中的表达及意义 被引量:3

EXPRESSION AND CLINICAL SIGNIFICANCE OF miR-181a-5p AND MEST IN TISSUE OF HIGH-GRADE SEROUS OVARIAN CANCER

摘要 目的探讨微小RNA 181a-5p(miR-181a-5p)及中胚层特异性转录基因(MEST)在高级别浆液性卵巢癌组织中的表达及临床意义。方法选取我院妇科2019年7月—2021年11月因高级别浆液性卵巢癌行手术治疗的患者56例,同期因子宫良性病变行附件切除的56例患者作为对照,采用实时荧光定量PCR技术(RT-qPCR)检测卵巢癌、正常卵巢及输卵管伞端组织中miR-181a-5p及MEST的表达。Pearson法分析卵巢癌组织中miR-181a-5p与MEST表达的相关性。卡方检验分析两者表达与患者肿瘤分期、有无淋巴结转移及血清中CA125水平等临床病理特征的关系。应用生物信息学软件预测miR-181a-5p与MEST之间的靶向结合位点,并应用双荧光素酶实验进一步验证。结果卵巢癌组织中miR-181a-5p、MEST的表达量与正常卵巢和输卵管伞端组织比较,差异有显著性(F=123.236、234.764,P<0.05)。Pearson相关性分析显示,卵巢癌组织中miR-181a-5p与MEST的表达呈显著负相关(r=-0.985,P<0.01)。miR-181a-5p与MEST的表达与患者肿瘤分期、有无淋巴结转移及血清中CA125水平等均显著相关(χ^(2)=4.548~33.547,P<0.05)。双荧光素酶实验显示,miR-181a-5p可抑制野生型MEST的荧光素酶活性(t=8.726,P<0.05),而对MEST突变型无影响(P>0.05),提示miR-181a-5p与MEST存在靶向结合位点。结论miR-181a-5p在高级别浆液性卵巢癌中表达下调,可能通过调控MEST表达在肿瘤进展及转移中发挥抑癌基因的功能。 Objective To investigate the expression and clinical significance of miR-181a-5p and the mesoderm-specific transcript gene(MEST)in the tissue of high-grade serous ovarian cancer.Methods This study included 56 patients who underwent surgery for high-grade serous ovarian cancer in the Department of Gynecology,The Affiliated Hospital of Qingdao University from July 2019 to November 2021,and selected 56 patients who underwent adnexectomy for benign uterine diseases during the same period as the control.Quantitative real-time PCR was used to measure the expression of miR-181a-5p and MEST in the ova-rian cancer,normal ovary,and the fimbri of the fallopian tube tissues.Pearson correlation analysis was performed to analyze the correlation between the expression of miR-181a-5p and MEST in the ovarian cancer tissue.The chi-squared test was used to analyze the relationship between their expression and clinical and pathological features(including tumor stage,lymph node metastasis,and serum CA125 level).Bioinformatic analysis was conducted to predict the target binding site between miR-181a-5p and the MEST gene,followed by verification with the dual luciferase assay.Results The expression of miR-181a-5p and MEST in the ovarian cancer tissues significantly differed from that in the tissues of normal ovary and the fimbriae of the fallopian tube(F=123.236,234.764,P<0.05).The Pearson correlation analysis showed that there was a significant negative correlation between the expression of miR-181a-5p and MEST in the ovarian cancer tissues(r=-0.985,P<0.01).The expression of miR-181a-5p and MEST was significantly associated with tumor stage,lymph node metastasis,and serum CA125 level(χ^(2)=4.548-33.547,P<0.05).The dual luciferase assay showed that miR-181a-5p inhibited the luciferase activity of the wild-type MEST luciferase repor-ter(t=8.726,P<0.05),but had no effect on the mutant MEST luciferase reporter(P>0.05),indicating the presence of a target binding site between miR-181a-5p and the MEST gene.Conclusion miR-181a-5p is downregulated in high-grade serous ovarian cancer,which may play an inhibitory role in tumor progression and metastasis by regulating the expression of the MEST gene.
