摘要
采用单因素试验及正交试验的方法对植物乳杆菌的高密度发酵配方和发酵条件进行优化,确定植物乳杆菌的最佳发酵培养基配方:胰蛋白胨12 g/L、牛肉膏8 g/L、酵母膏4 g/L、磷酸氢二钾2 g/L、柠檬酸三胺2 g/L、乙酸钠5 g/L、硫酸镁0.58 g/L、硫酸镁0.07 g/L、吐温80 1 mL/L、pH 6.5。最佳培养条件为起始pH 6.5、发酵温度35℃、接种量20 mL/L、溶氧量0 mL/L (控制低于100 mL/L)、搅拌转速50 r/min、罐压0.03 MPa。采用优化后的发酵参数,植物乳杆菌在10 L发酵罐中的菌体发酵密度可达9.63×10^(12)CFU/m L。
Single factor experiment and orthogonal experiment were used to optimize the formula and fermentation conditions of high density fermentation of Lactobacillus plantarum,and the optimal fermentation medium formula of Lactobacillus plantarum was determined as follows:tryptone 12 g/L,beef extract 8 g/L,yeast extract 4 g/L,dipotassium hydrogen phosphate 2 g/L,citrate triamine 2 g/L,sodium acetate 5 g/L,magnesium sulfate 0.58 g/L,magnesium sulfate 0.07 g/L,Tangu 80 1 mL/L and pH 6.4.The best culture condition were initial pH 6.5,fermentation temperature 35 ℃,inoculation amount 20m L/L,dissolved oxygen 0 mL/L(controlled below 100 mL/L),stirring speed 50 r/min and tank pressure 0.03 MPa.Using the optimized fermentation parameters,the fermentation density of L.plantarum in 10 L fermenter reached 9.63×10^(12)CFU/m L.
