摘要
目的:探讨清肤消肿散对急性皮下蜂窝组织炎大鼠的治疗效果及对TGF-β1/Smad信号通路的调控机制。方法:选取健康SD大鼠45只,随机分为正常对照组、模型对照组与研究组,其中模型对照组与研究组建立急性皮下蜂窝组织炎大鼠模型。通过酶联免疫吸附剂测定技术(ELISA)检测大鼠外周血中TGF-β1、IL-8和TNF-α水平;通过实时荧光PCR检测其皮下组织中TGF-β1、Smad2、Smad3、Smad7基因的mRNA水平;通过Western Blot法检测各组大鼠皮下组织中TGF-β1、Smad2、Smad3、Smad7基因的蛋白水平。结果:与正常对照组相比,模型对照组大鼠外周血中TGF-β1、IL-8和TNF-α水平显著上升(P<0.05);与模型对照组相比,研究组大鼠外周血中TGF-β1、IL-8和TNF-α水平显著降低(P<0.05)。与正常对照组相比,模型对照组大鼠皮下组织中TGF-β1、Smad2、Smad3基因的mRNA与蛋白水平显著上升,Smad7表达量明显降低(P<0.05);与模型对照组相比,研究组模型对照组大鼠皮下组织中TGF-β1、Smad2、Smad3基因的mRNA与蛋白水平显著降低,Smad7表达量明显增加(P<0.05)。结论:清肤消肿散能够通过调控TGF-β1/Smad信号通路治疗急性皮下蜂窝组织炎大鼠。
Objective:To investigate the therapeutic effect of Qingfu Xiaozhong powder on rats with acute subcutaneous cellulitis and TGF-β1/Smad signaling pathway.Methods:45 healthy SD rats were randomly divided into normal control group,model control group and study group.The model control group and study group established the rat model of acute subcutaneous cellulitis.The level of TGF-β1,IL-8,TNF-αin rat peripheral blood was detected by enzyme-linked immunosorbent assay(ELISA).mRNA levels of TGF-β1,Smad2,Smad3,Smad7 genes in subcutaneous tissue was detected by real-time fluorescent PCR.Protein levels of TGF-β1,Smad2,Smad3 and Smad7 genes in subcutaneous tissue of rats in each group was detected by Western blot.Results:Compared with the normal control group,the level of TGF-β1,IL-8,TNF-αin the peripheral blood of the model control group increased significantly(P<0.05).Compared with the model control group,the level of TGF-β1,IL-8,TNF-αin peripheral blood of rats in the study group decreased significantly(P<0.05).Compared with the normal control group,the mRNA and protein levels of TGF-β1,Smad2 and Smad3 genes in the subcutaneous tissue of the model control group increased significantly,and the expression of Smad7 decreased significantly(P<0.05).Compared with the model control group,the mRNA and protein levels of TGF-β1,Smad2 and Smad3 genes in the subcutaneous tissue of rats in the study group decreased significantly,and the expression of Smad7 increased significantly(P<0.05).Conclusion:Qingfu Xiaozhong powder can regulate TGF-β1/Smad signaling pathway in rats with acute subcutaneous cellulitis.
