大鼠过敏性猝死中HK-1的表达研究

摘要 目的:探究大鼠过敏性猝死后各时间段及各保存条件下HK-1的表达。方法:建立大鼠过敏性猝死模型,将50只大鼠分为对照组和实验组,再按照不同保存条件(常温、冷藏、冷冻)和时间段(0 h、24 h、48 h、7 d),将实验组分为0 h死亡组、24 h常温组、48 h冷藏组和7 d冷冻组,取其肺部和胃肠组织,分别用苏木素-伊红(Hematoxylin-Eosin,HE)染色、酶联免疫吸附测定(Enzyme-Linked Immunosorbent Assay,ELISA)和qRT-PCR法检测HK-1蛋白和m RNA的表达。结果:HE染色显示,各实验组肺部和胃肠组织有大量嗜酸性粒细胞浸润;ELISA和qRT-PCR检测结果显示,除了7 d冷冻组外,其余各实验组肺部和胃肠组织中HK-1蛋白和mRNA的表达均高于对照组(P<0.05)。结论:卵蛋白致大鼠过敏性猝死可引起肺部和胃肠组织HK-1表达升高,对48 h内过敏性猝死的法医学鉴定有参考价值。 Objective:To investigate the expression of HK-1 in various time periods and preservation conditions after sudden allergic death in rats.Methods:The model of sudden allergic death in rats was established.50 rats were divided into control group and experimental group.According to different storage conditions(normal temperature,cold storage,freezing)and time periods(0 h,24 h,48 h,7 d),the experimental group was divided into 0 h death group,24 h normal temperature group,48 h cold storage group and 7 d frozen storage group.The lung and gastrointestinal tissues were stained with hematoxylin eosin(HE),and the expression of HK-1 protein and mRNA was detected by enzyme-linked immunosorbent assay(ELISA)and qRT-PCR.Results:HE staining showed that there were a large number of eosinophil infiltration in the lung and gastrointestinal tissues of each experimental group.The results of ELISA and qRT-PCR showed that the expression of HK-1 protein and mRNA in the lung and gastrointestinal tissues of each experimental group were higher than those of the control group,except the 7d freezing group.Conclusion:sudden allergic death induced by ovalbumin can increase the expression of HK-1 in lung and gastrointestinal tissues,and it has reference value for the forensic identification of sudden allergic death within 48 hours.
