术连饮对腹泻型肠易激综合征模型大鼠脑肠肽的影响 被引量:1

摘要 目的探讨术连饮对腹泻型肠易激综合征(IBS-D)模型大鼠脑肠肽的影响。方法雄性SD大鼠40只,根据体重均衡原则,按照随机数表法分为空白组、模型组、匹维溴铵组和术连饮组,每组各10只。采用番泻叶灌胃联合束缚法建立IBS-D模型大鼠,观察大鼠体重、进食量、粪便Bristol评分变化。采用实时荧光定量PCR(RT-qPCR)法检测下丘脑和结肠组织P物质(SP)、神经肽Y(NPY)mRNA相对表达量,蛋白质印迹(Western blot)法检测SP、NPY蛋白表达水平。结果模型组大鼠体重、进食量均低于空白组,Bristol评分高于空白组,下丘脑和结肠组织SP蛋白及其mRNA表达均高于空白组,NPY蛋白及其mRNA表达均低于空白组;术连饮组大鼠体重、进食量均高于模型组,Bristol评分低于模型组,下丘脑和结肠组织SP蛋白及其mRNA表达均低于模型组,NPY蛋白及其mRNA表达均高于模型组,差异均有统计学意义(P<0.05)。结论术连饮能够改善IBS-D模型大鼠腹泻症状,其作用机制可能与调节下丘脑和结肠组织SP、NPY的表达有关。 Objective To investigate the impacts of Zhulian Yin on brain-gut peptides in rats with diarrhea-predominant irritable bowel syndrome(IBS-D).Methods A total of 40 male SD rats were randomly divided into the blank group,the model group,the pinaverium bromide group and the Zhulian Yin group according to the principle of body weight balance,with 10 rats in each group.The IBS-D rat model was established by gavage of folium sennae combined with binding method.The changes of body weight,food intake and stool Bristol score of rats were observed.The relative expressions of SP and NPYmRNA in hypothalamus and colon tissues were detected by real-time fluorescent quantitative PCR(RT-qPCR).The expression levels of substance P(SP)and neuropeptide(NPY)proteins were detected by Western blot.Results The body weight and food intake of rats in the model group were both lower than those in the control group.The Bristol score was higher than that in the control group.The SP protein and mRNA expressions in hypothalamus and colon tissues were both higher than those in the control group.The NPY protein and its mRNA expressions were both lower than those in the control group.The body weight and food intake of rats in the Zhulian Yin group were higher than those in the model group.The Bristol score was lower than that in the model group.The SP protein and its mRNA expressions in hypothalamus and colon tissues were both lower than those in the model group.The NPY protein and its mRNA expressions were both higher than those in the model group.The above differences were all statistically significant(P<0.05).Conclusion Zhulian Yin can improve the symptoms of diarrhea in IBS-D rat model,and its mechanism of action may be related to the regulation of SP and NPY expressions in the hypothalamus and colon tissues.
