摘要
为了解重组禽流感病毒(H5+H7)三价灭活苗在不同温度下对免疫效果的影响,本试验将160羽27日龄黄羽肉鸡随机分成4个组,每组40羽,分别注射不同温度重组禽流感病毒(H5+H7)三价灭活疫苗(H5N1 Re-11株+Re-12株,H7N9 H7-Re3株),评价其免疫抗体效价和免疫后不良反应。结果显示:接种29~31℃疫苗组受免鸡群应激反应较小,免疫28 d后剖检可见疫苗颗粒在注射部位及周边肌肉组织中分解吸收较好,免疫副反应优于其他温度组;首免14 d后4个组抗体均能达到免疫保护效价之上(>4log_(2)),4个组之间差异不显著(P>0.05);免疫28 d时,29~31℃组的抗体平均滴度均低于23℃以下各温度组,但仍保持在保护效价之上,且4个组之间差异极显著(P<0.01);首免28 d和二免28 d,H5N1 Re-11株+Re-12株,H7N9 H7-Re3株抗体可达到峰值水平,在抗体产生时间和消长规律上表现一致。结果表明重组禽流感病毒(H5+H7)三价灭活苗在6~31℃时能保证免疫效果,29~31℃时疫苗吸收效果较好,且注射部位肌肉内疫苗颗粒残留与疫苗注射温度呈反比,温度越高残留越少。
In order to assess the effect of recombinant avian influenza virus(H5+H7)trivalent inactivated vaccine on immune effect at different temperatures,this study randomly divided 160 broilers of 27-day-old into 4 groups,with 40 in each group.The broilers were injected with recombinant avian influenza virus(H5+H7)trivalent inactivated vaccine(H5N1 Re-11 strain+Re-12 strain,H7N9 H7-Re3 strain)at different temperatures,and their immune antibody titers and adverse reactions were evaluated.The results showed that the stress response of immunized chickens in the 29-31℃vaccine group was less,and after 28 days of immunization,the decomposition and absorption of vaccine particles in the injection site and surrounding muscle tissue were better,and the immune side effects were better than those in other temperature groups;14 days after the first immunization,the antibodies of the four groups reached above the immunoprotective titer(>4log_(2)),and no significant difference was observed among the four temperature groups(P>0.05);on the 28th day after immunization,the average titer of antibody in 29-31℃group was lower than that in each temperature group below 23℃,but it still remained above the protective titer,and the difference between each group was very significant(P<0.01);on the 28th day of the first immunization and the 28th day of the second immunization,the antibodies to H5N1 Re-11 strain+Re-12 strain,H7N9 H7-Re3 strain reached the peak level,and the antibody production time and growth and decline law were consistent.Thus,the trivalent inactivated vaccine of recombinant avian influenza virus(H5+H7)demonstrated the immune effect in the range of 6-31℃,the vaccine absorption effect was better at 29-31℃,and the vaccine particle residue in the muscle of the injection site was inversely proportional to the vaccine injection temperature,namely,the higher the temperature,the less the residue.
