摘要
目的:研究骨髓间充质干细胞(BMSCs)对大鼠牙槽骨缺损部分的再生修复作用,并探讨相关机制。方法:21只牙槽骨缺损大鼠随机分为模型组(10只)、BMSCs组(11只)。确认建模成功后BMSCs组于牙槽骨缺损处植入BMSCs膜片;模型组不植入膜片。干预后第3、6周Micro-CT活体扫描大鼠牙槽骨缺损处;HE染色观察骨和血管新生情况;Western blot法检测牙槽骨组织中抗酒石酸酸性磷酸酶(TRAP)蛋白表达。结果:与模型组比较,BMSCs组干预后第3、6周骨体积分数(BV/TV)、骨密度(BMD)增加,骨小梁分离度(Tb.sp)减少,TRAP蛋白表达降低(P<0.05);与干预后第3周比较,模型组、BMSCs组干预后第6周BV/TV、BMD增加,Tb.sp减少,TRAP蛋白表达降低(P<0.05)。结论:BMSCs可促进大鼠牙槽骨缺损部分的再生修复,推测其作用机制可能与下调TRAP蛋白表达从而抑制破骨细胞活性有关。
Objective:To investigate the effect of bone marrow mesenchymal stem cells(BMSCs)on the regeneration and repair of rat alveolar bone defects,and to explore the related mechanisms.Methods:21 rats with alveolar bone defect were randomly divided into model group(10)and BMSCs group(11).After successful modeling,the BMSCs group was implanted with a BMSCs membrane at the alveolar bone defect,and the model group was not implanted with a membrane.The alveolar bone defect in rats at 3 and 6 weeks after intervention was scanned by Micro-CT.The bone and angiogenesis were observed by HE staining method.Western blot was used to detect the expression of tartrate-resistant acid phosphatase(TRAP)protein in alveolar bone tissue.Results:Compared with the model group,the bone volume fraction(BV/TV)and bone density(BMD)of the BMSCs group was increased,the trabecular bone separation(Tb.sp)was decreased,and the expression of TRAP protein was decreased(P<0.05).Compared with the third week after intervention,BV/TV and BMD were increased,Tb.sp were decreased,and TRAP protein expression was decreased in the model group and BMSCs group at the sixth week after intervention(P<0.05).Conclusion:BMSCs can promote the regeneration and repair of rat alveolar bone defects.It is speculated that its mechanism may be related to down-regulating the expression of TRAP protein and inhibiting the activity of osteoclasts.
