生半夏秫米汤与法半夏秫米汤的急性毒性研究 被引量:3

Research on Acute Toxicity of Shengbanxia Shumi Decoction(生半夏秫米汤) and Fabanxia Shumi Decoction(法半夏秫米汤)

摘要 目的:探讨生半夏秫米汤与法半夏秫米汤的急性毒性。方法:120只SPF级昆明小鼠随机分为空白组、生半夏秫米汤组和法半夏秫米汤组,采用最大给药量试验,每日给药360 g/kg,连续观察14 d。实验第2、14天测血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、肌酐(CrE)、尿素氮(BUN)值和血常规,病理检查各脏器。结果:各组小鼠无死亡,血常规无用药相关异常变化。与空白组比较,生半夏秫米汤组和法半夏秫米汤组小鼠血清ALT、AST、CrE、BUN值均明显升高(P<0.05或P<0.01)。实验第2天,与生半夏秫米汤组比较,法半夏秫米汤组小鼠血清AST升高,ALT降低;实验第14天,与生半夏秫米汤组比较,法半夏秫米汤组小鼠血清ALT、AST、CrE、BUN均明显升高(P<0.01)。生半夏秫米汤组和法半夏秫米汤组小鼠肝肾病理学均见肝细胞水肿、点状坏死灶,肾小球扩张充血,肾小管上皮细胞肿胀,蛋白管型。法半夏秫米汤组小鼠肝细胞核溶解消失,肾小管变形及细胞管型;生半夏秫米汤组小鼠见再生肝细胞。结论:生半夏秫米汤和法半夏秫米汤有一定肝肾毒性,但毒性较低,法半夏秫米汤的损害更大。生半夏秫米汤造成的肝损害能否完全恢复,有待进一步研究。 Objective:To investigate the acute toxicity of Shengbanxia Shumi Decoction and Fabanxia Shumi Decoction.Methods:A total of 120 SPF Kunming mice were randomly divided into blank group,Shengbanxia Shumi Decoction group and Fabanxia Shumi Decoction group.The maximum dose test was performed,the mice were given the medicine once by gavage at the maximum dose of 360 g/kg and observed for 14 consecutive days.Serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),creatinine(CrE),blood urea nitrogen(BUN) and blood routine were measured on the 2ndand 14thday of the experiment.The pathological examination of organs were also performed.Results:There was no death in each group,and there was no abnormal change in blood routine.Compared with the blank group,the serum ALT,AST,CrE and BUN values of mice in the Shengbanxia Shumi Decoction group and the Fabanxia Shumi Decoction group were significantly increased(P <0.05 or P<0.01).On the 2ndday of the experiment,compared with the Shengbanxia Shumi Decoction group,the serum AST of the mice in the Fabanxia Shumi Decoction group was increased and ALT was decreased.On the14thday of the experiment,compared with the Shengbanxia Shumi Decoction group,the serum ALT,AST,CrE and BUN of the Fabanxia Shumi Decoction group were significantly increased(P<0.01).Liver and kidney pathology of mice in Shengbanxia Shumi Decoction group and Fabanxia Shumi Decoction group showed liver cell edema,spotty necrosis,glomerular dilatation and congestion,renal tubular epithelial cell swelling and protein tubular type.In the Fabanxia Shumi Decoction group,the nuclear dissolution of hepatocytes disappeared,renal tubular deformation and cell tubular type were observed.Regenerated hepatocytes were observed in mice in the Shengbanxia Shumi Decoction group.Conclusion:Shengbanxia Shumi Decoction and Fabanxia Shumi Decoction have certain hepatorenal toxicity,but the toxicity is low,and the damage of Fabanxia Shumi Decoction is greater.Whether the liver damage caused by Shengbanxia Shumi Decoction can be fully restored remains to be further studied.
