摘要
目的总结本研究室SD大鼠生育力与早期胚胎发育毒性试验中各指标的数值,建立相应指标的背景数据,为药物生殖毒理研究提供参考。方法从本研究室2012-2019年大鼠生育力与早期胚胎发育毒性试验中选取溶媒对照组相关指标的数据进行汇总,采用SPSS19.0统计软件进行正态性分布检验,计算x±s、95%可信区间和变异系数。溶媒对照组共484只SD大鼠,雌雄各半;交配后共有228对交配成功,219只雌性大鼠成功妊娠。相关指标包括交配前484只SD大鼠(雌雄各半)体重和摄食量、228只交配成功的雄性SD大鼠脏器和生育力指标(交配率、勃起潜伏期、交配成功时间和精子分析)及219只妊娠大鼠体重和早期胚胎发育指标(胎重、黄体数、活胎数、死胎数和吸收胎数)。结果交配前:雄性SD大鼠第1天(D_(1))体重(185±57)g,第1周(W_(1))摄食量每只(22.0±4.1)g·d^(-1),D_(60)体重(436±36)g,W_(9)摄食量每只(24.1±3.2)g·d^(-1);雌性SD大鼠D_(1)体重(202±19)g,W_(1)摄食量每只(14.9±1.8)g·d^(-1),D_(11)体重(221±17)g,W_(2)摄食量每只(16.0±1.8)g·d^(-1)。交配成功后:雄性SD大鼠附属生殖脏器重量分别为睾丸(2.912±0.250)g、附睾(1.003±0.110)g、包皮腺(0.076±0.036)g、前列腺(1.144±0.238)g、精囊腺(1.054±0.295)g、提肛肌(0.313±0.068)g和括约肌(1.081±0.156)g;交配率为94.2%,交配成功时间(2.6±1.5)d,阴茎勃起潜伏期(20.3±3.2)s;附睾精子数量(95.1±87.5)×10^(6)·g^(-1),活动率(56.5±18.8)%。妊娠大鼠妊娠第0天(GD_(0))体重(229±17)g,GD_(14)体重(295±19)g、妊娠率96.1%、平均黄体数(13.2±1.7)个、平均着床数(12.7±1.9)个、平均活胎数(12.0±2.1)只、活胎率94.8%、窝吸收胎率46.1%、窝死胎率3.7%和胎重(10.9±1.8)g。结论初步建立了本研究室SD大鼠生育力与早期胚胎发育毒性试验的背景数据,可为生殖毒性研究提供参考。
OBJECTIVE To summarize the data on studies of fertility and early embryo development toxicity in SD rats,and establish the related background data in our laboratory.METHODS From 2012 to 2019,the data on related indexes of the solvent control group during experiments on fertility and early embryo development toxicity in rats was collected and summarized.SPSS19.0 statistical software was used to test the normal distribution,while x±s,95%confidence interval and coefficient of variation were calculated.There were 484 SD rats in the solvent control group,half male and half female.After mating,228 pairs were successfully mated and 219 female rats became pregnant.The related indexes included the body mass and food consumption of 484 SD rats(half male and half female)before mating,the viscera and fertility indexes of 228 male SD rats that successfully mated(mating rate,penile erection latency period,time of successful mating and sperm analysis),and the body mass and early embryo development indexes of 219 pregnant rats(the fetal weight,amount of corpus luteum,number of live fetuses,number of dead fetuses and number of absorbed fetuses).RESULTS Before mating,the body mass on day 1(D_(1))and food consumption in week 1(W_(1))of male SD rats were(185.2±56.6)g and(22.0±4.1)g·d^(-1),respectively.The body mass on D_(60) and food consumption on W_(9) were(436.3±36.4)g and(24.1±3.2)g·d^(-1),respectively.The body mass in D_(1) and food consumption in W_(1) of female SD rats were(202.4±19.3)g and(14.9±1.8)g·d^(-1),respectively.The body mass on D_(11) and food consumption in W_(2) were(221.1±17.2)g and(16.0±1.8)g·d^(-1),respectively.After mating,the weight of accessory reproductive organs in male SD rats was(2.912±0.250)g for the testis,(1.003±0.110)g for the epididymis,(0.076±0.036)g for the preputial gland,(1.144±0.238)g for the prostate gland,(1.054±0.295)g for the seminal vesicle,(0.313±0.068)g for the levatorani muscle,and(1.081±0.156)g for the sphincter.The mating rate was 94.2%,and the successful mating time was(2.6±1.5)d.The penile erection latency period was(20.3±3.2)s.The sperm count of the epididymis was(95.1±87.5)×10^(6)·g^(-1),and the motility rate was(56.5±18.8)%.The body mass of pregnant rats was(229.3±16.8)g on gestation day 0(GD_(0)),and was(295.3±19.0)g on GD_(14).The rate of pregnancy was 96.1%,the average luteum number was(13.2±1.7),the average implantation number was(12.7±1.9),the average number of live fetuses was(12.0±2.1),the number live fetuses of rate was 94.8%,the fetal absorption rate(litter)was 46.1%,the fetal dead rate(litter)was 3.7%,and the gravid uterus mass was(10.9±1.8)g.CONCLUSION The background data on fertility and early embryo development toxicity in SD rats has been established in our laboratory,which can provide reference for reproductive toxicity studies.
