摘要
目的探讨CXC趋化因子受体7(CXCR7)和细胞周期数依赖性蛋白激酶4(CDK4)在骨肉瘤组织、骨软骨瘤组织的表达及临床意义。方法选择2018年1月至2022年1月齐齐哈尔医学院第五附属医院(大庆市龙南医院)、齐齐哈尔医学院附属第十医院(大庆油田总医院)和广东医科大学附属医院经病理学确诊的88例骨肉瘤组织标本和32例骨软骨瘤组织标本,采用免疫组织化学法检测CXCR7和CDK4在上述组织中的表达,采用χ2检验行统计学分析,分析CXCR7和CDK4的表达与骨肉瘤各临床病理因素之间的关系。结果骨肉瘤组织CXCR7表达率为72.73%(64/88),明显高于骨软骨瘤组织CXCR7表达率[18.75%(6/32)],两者差异有统计学意义(χ2=28.130,P<0.01)。CXCR7表达水平与骨肉瘤患者恩内金分期(Eneeking分期)、肺转移、软组织浸润明显相关(χ2=4.982、6.094、5.029,P<0.05),CXCR7表达水平与骨肉瘤患者性别、年龄、组织类型、病变部位、碱性磷酸酶(ALP)水平无相关(χ2=0.008、0.008、0.139、0.002、0.071,P>0.05)。骨肉瘤组织CDK4表达率为72.73%(60/88),明显高于骨软骨瘤组织CDK4表达率[18.75%(6/32)],两者差异有统计学意义(χ2=9.205,P<0.01)。CDK4表达水平与骨肉瘤患者Eneeking分期、肺转移明显相关(χ2=5.430、4.897,P<0.05),CDK4表达水平与骨肉瘤患者性别、年龄、组织类型、病变部位、软组织浸润、ALP水平无相关(χ2=0.048、0.045、0.091、0.012、1.056、0.282,P>0.05)。结论CXCR7和CDK4过度表达是骨肉瘤侵袭、转移的生物学标志,联合检测CXCR7和CDK4有助于评估骨肉瘤生物学特性。
Objective To Investigate the expression and clinical significance of CXC chemokine receptor 7(CXCR7)and cell cycle number dependent protein kinase 4(CXCR7)in osteosarcoma tissues and osteochondroma tissues.Methods Totally,88 osteosarcoma tissue samples and 32 osteochondroma tissue samples were collected from the Fifth Affiliated Hospital of Qiqihar Medical College(Daqing Longnan Hospital),the Tenth Affiliated Hospital of Qiqihar Medical College(Daqing Oilfield General Hospital)and Affiliated Hospital of Guangdong Medical University from January 2018 to January 2022.Immunohistochemistry was used to detect the expression of CXCR7 and CDK4 in the above tissues.Chi-square test was used for statistical analysis to analyze the relationship between CXCR7 and CDK4 expression and various clinicopathological factors of osteosarcoma.Results The CXCR7 expression rate in osteosarcoma tissues was 72.73%(64/88),which was significantly higher than that in osteochondroma tissues[18.75%(6/32),χ2=28.130,P<0.01].The expression of CXCR7 was correlated osteosarcoma patients with Eneeking stage,lung metastasis and soft tissue infiltration(respectively,χ2=4.982,6.094,5.029,P<0.05).The expression of CXCR7 was not significantly correlated with gender,age,tissue type,lesion site and alkaline phosphatase(ALP)level in osteosarcoma patients(respectively,χ2=0.008,0.008,0.139,0.002,0.071,P>0.05).The expression CDK4 rate in osteosarcoma tissues was 72.73%(60/88),which was significantly higher than that in osteochondroma tissues[18.75%(6/32),χ2=9.205,P<0.01].The expression of CDK4 was correlated with Eneeking stage and lung metastasis in osteosarcoma patients(χ2=5.430,4.897,P<0.05),but not with gender,age,tissue type,lesion site,soft tissue infiltration and ALP level(respectively,χ2=0.048,0.045,0.091,0.012,1.056,0.282,P>0.05).Conclusion The overexpression of CXCR7 and CDK4 might be important biological markers for invasion and metastasis of osteosarcoma,and the combined detection of CXCR7 and CDK4 might be the predictors for biological evaluation of osteosarcoma.
