摘要
目的探讨白细胞介素-38中和抗体(IL-38 nAb)对肝癌细胞生长的影响及其机制。方法30只Balb/c小鼠随机分成对照组和IL-38 nAb组,通过肝脏右外叶包膜下注射小鼠肝癌细胞H22建立肝癌模型,IL-38 nAb组小鼠每2天腹腔注射IL-38 nAb(50μg/只),对照组给予等量的生理盐水。10 d后处死小鼠,统计肿瘤大小和肝脏质量,检测肝癌组织Bax和Bcl-2表达情况;检测小鼠血清IL-1β、IL-4、IL-10和IL-12含量;检测小鼠肿瘤相关巨噬细胞(TAM)的表达情况;检测小鼠肿瘤组织诱导型一氧化氮合酶(iNOS)、IL-1β、精氨酸酶-1(Arg-1)和IL-10 mRNA表达。结果与对照组比较,IL-38 nAb组小鼠肝癌组织大小和肝脏质量均明显降低,而Bax/Bcl-2表达明显升高(P<0.05);与对照组相比,IL-38 nAb组小鼠肝癌组织中TAM比例明显降低,M1/M2比值、iNOS和IL-1βmRNA表达升高,Arg-1和IL-10 mRNA表达降低(P<0.05);与对照组相比,IL-38 nAb组小鼠血清IL-1β和IL-12表达明显升高(P<0.05),IL-4和IL-10表达明显降低(P<0.05)。结论IL-38 nAb可以抑制肝癌细胞生长,机制可能与TAM极化有关。
To investigate the role of interleukin-38(IL-38)neutralizing antibody(IL-38 nAb)in the development of liver cancer,30 Balb/c mice were recruited and randomly divided into the control group and IL-38 nAb group.The liver cancer model was established by subcapsular injection of mouse hepatocarcinoma cells H22 in the right outer lobe of the liver.The mice in the IL-38 nAb group were intraperitoneally injected with IL-38 nAb(50μg/mouse)every two days,and sacrificed 10 days later to count the tumor size and liver mass.The expression of Bax and Bcl-2 in liver cancer tissues,and serum IL-1β,IL-4,IL-10 and IL-12 levels were detected;the expression of tumor-associated macrophages(TAM)and the expression of iNOS,IL-1β,Arg-1 and IL-10 mRNA were detected;compared with the control group,the liver cancer tissue size and liver mass of mice in the IL-38 nAb group were significantly reduced,while the expression of Bax/Bcl-2 in the IL-38 nAb group was significantly increased(P<0.05);compared with the control group,the TAM expression and the expression of Arg-1 and IL-10 mRNA in the IL-38 nAb group were significantly reduced,and the ratio of M1/M2 expression and the expression of iNOS and IL-1βmRNA were significantly increased(P<0.05);compared with the control group,serum IL-1βand IL-12 expressions in the IL-38 nAb group were significantly increased,and IL-4 and IL-10 expressions were significantly decreased(P<0.05).Taken together,this study uncovers that IL-38 nAb can inhibit the growth of liver cancer by regulating the polarization of TAM.
