摘要
目的探讨杞黄降糖胶囊对糖尿病模型大鼠血糖及胰岛功能的影响,及其对胰腺病理学变化的影响。方法取SD大鼠80只,雌雄各半,随机选取10只作为正常组,其余大鼠舌下静脉注射四氧嘧啶(45 mg/kg)制作糖尿病大鼠模型。最终50只成模大鼠根据血糖、性别、体质量分为5组:每组10只,模型+盐水对照组(20 mL/kg);模型+消渴丸(1.25 g/kg)组;模型+杞黄降糖胶囊大剂量(3.48 g/kg)组;模型+杞黄降糖胶囊中剂量(1.74 g/kg)组;模型+杞黄降糖胶囊小剂量(0.88 g/kg)组。连续灌胃给药10 d,测定大鼠一般体征指标,检测血糖、胰岛素等指标,HE染色观察胰腺组织的病理变化。结果与正常组相比,模型组大鼠血糖水平明显升高(P<0.01)。与模型+盐水组比较,模型+消渴丸组大鼠给药后血糖水平明显降低(P<0.01);模型+杞黄降糖胶囊大、中剂量组血糖水平降低明显(P<0.05);杞黄降糖胶囊小剂量组血糖降低不明显(P>0.05)。与模型+盐水组比较,模型+杞黄降糖胶囊大、小剂量组的糖耐量曲线下面积均明显降低,且两组多数时间点的血糖水平低于模型盐水组。模型+消渴丸和模型+杞黄降糖胶囊大、中剂量组糖耐量曲线下面积与模型+盐水组比较均明显降低(P<0.05)。服糖30 min后,模型+消渴丸组动物胰岛素水平明显升高;杞黄降糖胶囊大、中剂量组与服糖前比较,胰岛素水平显著升高。胰岛病理结果显示模型+杞黄降糖胶囊治疗各组的胰腺组织较模型+盐水组比较,其受损胰腺组织结构均有所改善,且杞黄降糖胶囊大剂量组明显。结论杞黄降糖胶囊能促进高糖状态下大鼠胰岛素分泌,对糖尿病大鼠有明显降低血糖的作用,对四氧嘧啶造成的大鼠胰腺组织损伤有一定的修复作用。
Objective To investigate the effect of Qihuang Jiangtang Capsule(杞黄降糖胶囊)on blood glucose and pancreatic islet function in diabetic rats.Methods Eighty SD rats with half male and half female were selected.Ten rats were randomly se-lected as the normal group and the rest rats were injected tetraoxypyrimidine(45 mg/kg)into the sublingual vein to make the dia-betic rats.The final 50 model rats were divided into 5 groups according to blood glucose,sex and body mass,10 rats in each group:model+saline control group(20 mL/kg),model+Xiaoke Pill(消渴丸)group(1.25 g/kg),model+Qihuang Jiangtang Capsule high dose group(3.48 g/kg),model+Qihuang Jiangtang Capsule medium dose group(1.74 g/kg),model+Qihuang Jiangtang Capsule low dose(0.88 g/kg).The drug was administered for 10 days.The general signs of rats,the levels of blood glucose and insulin were measured.The pathological changes of pancreatic tissue were observed by HE staining.Results Com-pared with that of the normal group,the blood glucose level of the rats in the model group was significantly increased(P<0.01).Compared with that of the model+saline group,the blood glucose level of the rats in the model+Xiaoke Pill group was signifi-cantly lower after administration(P<0.01),and the blood glucose level of the model+Qihuang Jiangtang Capsule high and me-dium dose groups was significantly lower(P<0.05),the blood glucose level of the Qihuang Jiangtang capsule low dose group was not significantly reduced(P>0.05).Compared with that of the model+saline group,the area under the glucose tolerance curve of the model+Qihuang Jiangtang capsule high and low dose groups was significantly decreased,and the blood glucose levels of the two groups at most time points were lower than those of the model+saline group.The area under the glucose tolerance curve of the model+Xiaoke Pill group and the model+Qihuang Jiangtang Capsule high and medium dose groups was significantly lower than that of the model+saline group(P<0.05).After taking sugar for 30 minutes,the insulin level of the animals in the model Xiaoxie Pill group was significantly increased.Compared with that of the Qihuang Jiangtang Capsule high and medium dose groups before taking sugar,the insulin level was significantly increased.The results of islet pathology showed that the pancreatic tissue structure of model+Qihuang Jiangtang Capsule high,medium and low dose groups was improved compared with that of the model+saline group,and that of the Qihuang Jiangtang Capsule high dose group was significantly improved.Conclusion Qihuang Jiangtang Capsule can promote the insulin secretion in rats under high glucose state,significantly reduce the blood glucose level in diabetic rats,and has a certain repair effect on pancreatic tissue damage caused by tetraoxypyrimidine.
