利用斑马鱼模型评价三消草不同提取物的抗癫痫活性 被引量:10

Study on anti-epileptic activity of Trifolium repens L.extracted by different methods on zebrafish model

摘要 目的:本研究旨在明确三消草的抗癫痫活性并比较不同提取方法的三消草缓解癫痫作用效果。方法:通过建立斑马鱼癫痫模型,采用行为学实验方法,分别对不同提取方法的三消草抗癫痫活性进行综合对比,并以丙戊酸钠为阳性对照,进一步明确三消草有效提取物的抗癫痫效果。应用实时荧光定量聚合酶链式反应(quantitative real-time polymerase chain reaction,qRT-PCR)检测癫痫标志性基因c-fos的表达水平,拟验证三消草的抗癫痫活性。结果:综合行为学运动力和活动力的检测结果,筛选出三消草总水提物、脱脂水提物和水提醇沉上清液具有抗癫痫活性。qRT-PCR验证结果与行为学一致。结论:本研究建立了一种联合运动力和活动力的行为学检测方法,用于检测中药的抗癫痫活性。通过生化指标进一步验证,明确了三消草的抗癫痫活性。综合各项指标,三消草的总水提物抗癫痫效果最佳。 Objective:To explore the anti-epileptic activity of Trifolium repens L.and compare the effects of Trifolium repens L.extracted by different methods on relieving epilepsy.Methods:Zebrafish epilepsy model was established and the behavioral assays were performed to compare the anti-epileptic activity of Trifolium repens L.extracted by different methods.In addition,sodium valproate was used as the positive control to further clarify the anti-epileptic effect of Trifolium repens L.Quantitative Real-time PCR was used to detect the expression levels of epilepsy marker gene c-fos,verifying the anti-epileptic activity of Trifolium repens L.Results:It was found that total water extract,defatted water extract,and water extraction and alcohol precipitation supernatant of Trifolium repens L.had anti-epileptic activity as shown by comprehensively analyzing the locomotion results.The quantitative real-time PCR results were consistent with the locomotion findings.Conclusion:In this study,a behavioral method combining locomotor capacity and mobility was established,which can be used to detect the anti-epileptic activity of traditional Chinese medicine.Results got from biochemical indicator assays further verified the anti-epileptic activity of Trifolium repens L.Based on the comprehensive results of various indicators,the total water extract has the best anti-epileptic activity.
