HPLC法同时测定白喉乌头中双酯型二萜生物碱含量

Simultaneous Determination of Diester Diterpene Alkaloids in Aconitum diphtheriae by HPLC

摘要 采用高效液相色谱法测定白喉乌头中的乌头碱、新乌头碱、次乌头碱的含量。采用Agilent C_(18)柱(4.6mm×150 mm,5μm),流动相为甲醇-水(20︰80),流速0.8 mL/min,检测波长235 nm,柱温30℃,进样量10μL。结果表明:新乌头碱、乌头碱和次乌头碱分别在1~5,0.35~1.75和0.5~2.5μg范围内呈良好线性关系;精密度S_(RSD)=0.42%(n=5);新乌头碱表明溶液在24 h内基本稳定,S_(RSD)=1.1%(n=5);新乌头碱重现性为S_(RSD)=0.97%(n=5);乌头碱平均含量0.2203 mg/g,S_(RSD)=0.67%(n=5);加标回收率新乌头碱为98.6%,S_(RSD)=0.85%(n=5),乌头碱为99.0%,S_(RSD)=0.93%(n=5)次乌头碱为98.7%,S_(RSD)=0.98%(n=5)。该方法简便、可靠、准确,可用于附子和白喉乌头中新乌头碱、乌头碱、次乌头碱的含量测定,为全面开发利用附子和白喉乌头药材提供进一步依据。 To determine the contents of aconitine,neoaconitine and hypaconitine in Diphtheria aconitum by HPLC,Agilent C_(18) column(4.6 mm×150 mm,5μm)was used,with mobile phase methanol water(20︰80),flow rate 0.8 mL/min,detection wavelength 235 nm,column temperature 30℃ and injection volume 10μL.The result showed that neoaconitine,aconitine and hypoaconitine showed good linear relationship in the range of 1-5,0.35-1.75 and 0.5-2.5μg respectively.The precision was S_(RSD)=0.42%(n=5);Neoaconitine showed that the solution was basically stable within 24 h,S_(RSD)=1.1%(n=5).The reproducibility of neoaconitine was S_(RSD)=0.97%(n=5).The average content of aconitine was 0.2203 mg/g,with S_(RSD)=0.67%(n=5).The recoveries of neoaconitine,aconitine and hypoaconitine were 98.6% with S;=0.85%(n=5),99.0% with S_(RSD)=0.93%(n=5),and 98.7%with S_(RSD)=0.98%(n=5).The method is simple,reliable and accurate.It could be used for the determination of neoaconitine,aconitine and hypaconitine in aconite and diphtheria Aconitum,and provides further basis for the comprehensive development and utilization of aconite and diphtheria Aconitum.
