摘要
【目的】筛选珍珠龙胆石斑鱼(Epinephelusfuscoguttatus♀×Epinepheluslanceolatus♂)豆粕诱导型肠炎(SBMIE)肠道组织的潜在生物标志物。【方法】在基础饲料中用豆粕替代质量分数0%、20%和40%的鱼粉蛋白(分别记为FM、SBM20和SBM40组),配制3种等氮(粗蛋白质量分数50%)等脂(粗脂肪质量分数10%)饲料,饲喂初始体质量约9 g的珍珠龙胆石斑鱼10周,分析石斑鱼生长性能、肠道组织结构、炎性指标和肠炎表征;通过非靶向代谢组学技术,筛选石斑鱼SBMIE肠道组织潜在生物标志物。【结果】实验水平豆粕替代鱼粉对珍珠龙胆石斑鱼的增重率、特定生长率和肝体指数有显著负面影响(P <0.05);对存活率无显著影响(P> 0.05);免疫指标和H&E组织切片显示,肠炎症状随SBM增多逐渐加重。SBM20和SBM40组中分别发现17和18种SBMIE潜在生物标志物,其中有13种核心生物标志物为两组共有,包括染料木素、大豆黄酮、染料木素-4’-O葡萄糖醛酸酯、鸡黄豆素、柠檬苦素、吲哚-3-丙烯酰甘氨酸、白三烯B4二甲胺、维生素C、黄芩苷、葡萄糖二酸、大豆皂苷Ⅰ、小肽(Leu-Gly-Pro)和戊基-4-羟脯氨酸。所有标志物中,除柠檬苦素、吲哚-3-丙烯酰甘氨酸、Leu-Gly-Pro、戊基-4-羟脯氨酸、癸二酸、磷酸肌酸、N,N-二甲基甲氧基噻吩、N-甲基蒽酰胺和2-氨基-3,4-二甲基咪唑(4,5-F)喹啉与FM组比强度显著降低外(P <0.05),其余强度均显著升高(P <0.05)。大部分标志物间有显著正相关或负相关作用(P <0.05)。【结论】在SBMIE状态下,珍珠龙胆石斑鱼肠道组织中存在特征性的潜在核心生物标志物。
【Objective】This study was designed to screen the potential biomarkers for soybean meal induced enteritis(SBMIE) of pearl gentian grouper, Epinephelus fuscoguttatus♀ × Epinephelus lanceolatus♂.【Method】Soybean meal(SBM) was used to replace 0%, 20% and 40%(mass fraction) of fish meal protein(recorded as FM, SBM20 and SBM40 groups, respectively) in the basal diets. Three kinds of equal nitrogen(50% crude protein) and equal fat(10% crude fat) diets were prepared to feed pearl gentian grouper with an initial weight of about 9 g for 10 weeks. Then, the effects of replacement of FM by SBM on the growth performance, intestinal tissue structure, inflammatory indexes and enteritis of pearl gentian grouper were analyzed.【Results】At the experimental level, The replacement of FM by SBM had significant negative effects on the weight gain rate, specific growth rate and hepatosomatic index of pearl gentian grouper(P < 0.05), but had no significant effect on survival rate(P > 0.05);the immune and histology analysis of intestine showed that the enteritis symptoms gradually aggravated with the increase of SBM. 17 and 18 metabolites were identified as potential biomarkers of SBMIE in SBM20 and SBM40groups, respectively. Among them, 13 core biomarkers coexist in two groups, including Genistein,daidzein, enistein 4’-O-glucuronide, olmelin, limonin, Indolyl-3-acryloylglycine, leukotriene B4dimethylamide, ascorbic acid, baicalin, D-saccharic acid, soyasaponin I, Leu-Gly-Pro,Valyl-4-hydroxyproline. The intensities of limonin, indolyl-3-acryloylglycine, Leu-Gly-Pro,valyl-4-hydroxyproline, sebacic acid, phosphocreatine, N, N-didesmethyltamoxifen, N-methylanthranilamide and 2-amino-3,4-dimethylimidazo(4,5-F) quinoline significantly decreased compared with those in FM group(P < 0.05), the intensities of other biomarkers significantly increased(P < 0.05). Most of the biomarkers had significant positive or negative correlation(P < 0.05).【Conclusion】There are characteristic potential core biomarkers in the intestinal tissue of pearl gentian groupers suffering from SBMIE.
