摘要
本试验旨在探究饲粮途径联合重金属暴露[镉(Cd)、铅(Pb)、汞(Hg)、铬(Cr)]对蛋鸡肝脏、肾脏、胫骨及血清中金属离子稳态的毒性及硒(Se)的缓解效果。试验选取63周龄健康罗曼粉蛋鸡160只,随机分为4个组,每组10个重复,每个重复4只鸡。对照组(CON组)饲喂玉米-豆粕型基础饲粮,加硒组(Se组)在基础饲粮中添加0.4 mg/kg Se(酵母硒),重金属联合暴露组(HEM组)基础饲粮中添加5 mg/kg Cd(氯化镉)、5 mg/kg Cr(氯化铬)、50 mg/kg Pb(硝酸铅)和3 mg/kg Hg(氯化汞),重金属联合暴露加硒组(HEM+Se组)在HEM组饲粮基础上添加0.4 mg/kg Se(酵母硒)。预试期1周,正试期12周。试验结束后,每个重复随机选取2只蛋鸡,翅静脉采血后屠宰,收集肝脏、肾脏、胫骨样品,测定Cd、Pb、Hg、Cr、Se、铜(Cu)、铁(Fe)、锰(Mn)、锌(Zn)、钠(Na)、钾(K)、镁(Mg)、钙(Ca)含量。结果表明:1)与CON组相比,HEM组肝脏、肾脏、胫骨中Cd、Pb、Hg含量显著升高(P<0.05)。与HEM组相比,HEM+Se组肝脏中Cd、Pb含量无显著差异(P>0.05),肾脏中Cd、Pb、Hg含量无显著差异(P>0.05),胫骨中Cd、Hg含量无显著差异(P>0.05)。2)与CON组相比,HEM组肝脏中Fe、Zn、K含量显著升高(P<0.05),血清中Se、Mn、Mg、Ca含量显著下降(P<0.05),胫骨中Se、Na、K含量显著升高(P<0.05)。与HEM组相比,HEM+Se组肝脏中Fe、Zn、K含量无显著差异(P>0.05),血清中Mn、Mg、Ca含量无显著差异(P>0.05),胫骨中Mn含量无显著差异(P>0.05)。3)相关性分析结果表明,蛋鸡肝脏、肾脏、胫骨及血清中13种金属离子含量之间依次存在19种、12种、7种和12种显著的相关关系(P<0.05)。由此可见,在本试验条件下,饲粮途径联合重金属暴露能引起组织金属离子紊乱,增加相应重金属的沉积,扰乱蛋鸡肝脏、肾脏、血清、胫骨中金属离子稳态,饲粮中添加0.4 mg/kg Se(酵母硒)未能缓解重金属暴露引起的金属离子紊乱毒性。
The objective of this experiment was to explore the dietary co-treatment of heavy metal[cadmium(Cd),lead(Pb),mercury(Hg)and chromium(Cr)]on metal ion homeostasis toxicity in liver,kidney,serum and tibia of laying hens,as well as the ameliorative effect of selenium(Se). A total of 160 healthy 63-week-old Lohmann pink-shell laying hens were randomly divided into 4 groups with 10 replicates per group and10 hens per replicate. Hens in the control group(CON group)were fed a corn-soybean meal basal diet,hens in the adding selenium group(Se group)were fed the basal diet supplemented with 0.4 mg/kg Se(selenium yeast),hens in the heavy metal co-treatment group(HEM group)were fed the basal diet supplemented with5 mg/kg Cd(CdCl_(2)),5 mg/kg Cr(CrCl_(3)),50 mg/kg Pb[Pb(NO_(3)) _(2) ]and 3 mg/kg Hg(HgCl2),and hens in the heavy metal co-treatment adding selenium group(HEM+Se group)were fed the HEM group diet supplemented with 0.4 mg/kg Se(selenium yeast). The pre-experimental period lasted for 1 week,and the experimental period lasted for 12 weeks. At the end of the experiment,two hens were selected from each replicate,the blood was collected from pterygoid vein and then slaughtered,the liver,kidney and tibia samples were collected,and the contents of Cd,Pb,Hg,Cr,Se,copper(Cu),iron(Fe),manganese(Mn),zinc(Zn),sodium(Na),potassium(K),magnesium(Mg)and calcium(Ca)were measured. The results showed as follows:1)compared with the CON group,the contents of Cd,Pb and Hg in liver,kidney and tibia were significantly increased(P<0.05). Compared with the HEM group,the contents of Cd and Pb in liver of HEM+Se group were no significant difference(P>0.05),the contents of Cd,Pb and Hg in kidney were no significant difference(P>0.05),and the contents of Cd and Hg in tibia were no significant difference(P>0.05). 2)Compared with the CON group,the contents of Fe,Zn and K in liver were significantly increased(P<0.05),the contents of Se,Mn,Mg and Ca in serum were significantly decreased(P<0.05),and the contents of Se,Na and K in tibia were significantly increased(P<0.05). Compared with the HEM group,the contents of Fe,Zn and K in liver of HEM+Se group were no significant difference(P>0.05),the contents of Mn,Mg and Ca in serum were no significant difference(P>0.05),and the tibia Mn content was no significant difference(P>0.05). 3)The correlation analysis results showed that there were 19,12,7 and 12 significant correlations among the contents of 13 metal ions in liver,kidney,tibia and the serum of laying hens(P<0.05),respectively. In summary,under the condition of this experiment,dietary co-treatment of heavy metal can induce the tissue metal ion disorder,and increase the corresponding heavy metal deposition,and disturb the metal ion homeostasis in liver,kidney,serum and tibia of laying hens. Dietary 0.4 mg/kg Se dose not alleviate the metal ion disorder toxicity cause by heavy metal treatment.
