γ-谷维素和半胱胺对应激小鼠氧化还原状态及免疫功能的影响 被引量:6

摘要 为探讨γ-谷维素和半胱胺对应激小鼠氧化还原状态和免疫功能的影响,将90只5周龄雄性ICR小鼠随机分为6组,每组15只,采用糖皮质激素诱导的小鼠应激模型,同时在饲料中分别添加不同剂量的γ-谷维素和半胱胺进行给药,饲养8周。结果表明:与模型组相比,联用1组的组织总SOD活力、血浆和组织GSH-Px活力和T-AOC均显著提高(P<0.05),血浆和组织MDA水平均显著降低(P<0.05);联用1组线粒体Mn-SOD活力、膜电位、乙酰辅酶A和ATP水平均显著提高(P<0.05),线粒体MDA质量摩尔浓度均显著降低(P<0.05);联用1组脾脏指数、胸腺指数和血浆中免疫球蛋白质量浓度均显著提高(P<0.05),TNF-α和IL-6质量浓度均显著降低(P<0.05),IL-10质量浓度显著提高(P<0.05)。实验结果显示,4个用药组对应激小鼠的氧化还原状态和免疫功能均有一定的改善作用,其中联用1组效果最显著,说明当γ-谷维素与半胱胺以较低剂量联用时可以发挥协同作用,起到很好的抗氧化和调节免疫功能的作用,从而有效对抗氧化应激带来的危害。 The effects of γ-oryzanol and cysteamine on the redox status and immunity of stressed mice were investigated. Ninety male ICR mice aged five weeks were randomly divided into 6 groups with 15 mice in each group. Stressed mouse model was developed by glucocorticoid injection,meanwhile, different doses of γ-oryzanol and cysteamine were added to the feed for 8-week administration. Compared with the model group, the activity of total superoxide dismutase(SOD) in tissues, the activity of glutathione peroxidase(GSH-Px) and the total antioxidant capacity(T-AOC) in plasma and tissues of the combination group 1 were significantly increased(P<0.05), while the malondialdehyde(MDA) content in plasma and tissues was significantly decreased(P<0.05). The activity of Mn-SOD, membrane potential, acetyl coenzyme A(A-CoA), and ATP in the mitochondria of combination group 1 were significantly increased(P<0.05), while the mitochondrial MDA content was significantly decreased(P<0.05). The spleen index, thymus index and the content of immunoglobulin in the plasma of combination group 1 were significantly increased(P<0.05). The levels of tumor necrosis factor-α(TNF-α) and interleukin-6(IL-6) were significantly decreased(P<0.05), while the level of interleukin-10(IL-10) was significantly increased(P<0.05). The results showed that the redox status and immune function of stressed mice were improved in the four experimental groups, and the combination group 1 had the most significant effect, indicating that the combination of γ-oryzanol and cysteamine at a lower dose could play a synergistic role in anti-oxidation and regulating immune function, thus effectively resisting the harm caused by oxidative stress.
