摘要
目的:探讨维生素D结合蛋白基因(GC基因)单核苷酸多态性与海南汉族过敏性鼻炎(AR)的易感性分析。方法:选取2017年—2020年海南省海口市某医院耳鼻喉头颈外科就诊的88例AR患者作为实验组(AR组)和同期在该院体检科体检的105例健康人群作为对照组(HC组),利用Snapshot技术检测GC基因三个SNP位点rs222016、rs4588、rs2282679的基因型,并通过人工、Snpstats云分析平台、SPSS25.0软件进行Hardy-weinberg平衡验证及基因型、等位基因、单倍型等进行统计学分析,对基因型有统计学差异的多态性位点进行logistic回归分析。结果:三个多态性位点rs222016、rs4588、rs2282679均可检测出三种基因型,其中rs222016基因型为GG、GA、AA,等位基因为G、A。rs4588基因型为AA、AC、CC,等位基因为A、C。rs2282679基因型为AA、AC、CC,等位基因为A、C。其中rs4588和rs2282679基因型及等位基因在组间的分布频率,差异无统计学意义(χ^(2)=0.101、0.523,P>0.05),rs222016基因型、等位基因在组间的分布频率,差异有统计学意义(χ^(2)=6.565,P<0.05),通过logsitic回归分析发现,等位基因G为保护因素。将这三个位点进行连锁分析发现它们构成的单倍型rs222016-rs4588-rs2282679-A-C-A与海南变应性鼻炎,差异有统计学意义(P<0.05)。结论:GC基因的多态性位点rs222016的多态性与海南汉族AR易感性有关,位点rs4588、rs2282679的多态性与海南汉族AR易感性无关,但这两个多态性位点的单倍型可协同增强位点rs222016对海南汉族AR的易感性。
Objective:To explore the susceptibility analysis of vitamin D binding protein gene(GC gene)single nucleotide polymorphism and allergic rhinitis(AR)in Hainan Han population.Methods:A total of 88 patients with AR who visited the hospital from 2017 to 2020 were selected as the experimental group(AR group),and 105 healthy people who received physical examination in the hospital during the same period were selected as the control group(HC group).Snapshot technology was used to detect the genotypes of the three SNP loci rs222016,rs4588,and rs2282679 of the GC gene.And through manual,Snpstats cloud analysis platform and SPSS 25.0 software Hardy-weinberg balance verification was done,and genotype,allele,haplotype,etc.were statistically analyzed.The polymorphic loci with statistically different genotypes were analyzed by logistic regression.Results:Three genotypes could be detected at the three polymorphic sites rs222016,rs4588,and rs2282679.Among them,rs222016 genotypes were GG,GA,AA,and alleles were G,A.The genotypes of rs4588 were AA,AC,and CC,and the alleles were A and C.The genotypes of rs2282679 were AA,AC,and CC,and the alleles were A and C.There was no statistically significant difference in the distribution frequency of rs4588 and rs2282679 genotypes and alleles between groups(χ^(2)=0.101,0.523,P>0.05).There was statistically significant difference in the distribution frequency of rs222016 genotype and alleles between the groups(χ^(2)=6.565,P<0.05).Log-sitic regression analysis found that allele G was a protective factor.Linkage analysis of these three loci found that there was statistically significant difference between their haplotype rs222016-rs4588-rs2282679-A-C-A and allergic rhinitis in Hainan(P<0.05).Conclusion:The genetic variation of GC gene polymorphism locus rs222016 is related to the susceptibility of Hainan AR,and the genetic variation of rs4588 and rs2282679 is not related to the susceptibility of AR,but the haplotype of these two polymor-dial sites can synergistically enhance the susceptibility of rs222016 to AR in Hainan.
