芪白平肺胶囊对COPD痰瘀阻肺证大鼠肺血管Bax、Bcl-2表达的影响 被引量:8

摘要 目的:研究芪白平肺胶囊改善COPD痰瘀阻肺证大鼠肺血管重构与增殖/凋亡相关分子Bax/Bcl-2的关系。方法:雄性SD大鼠随机分为正常组、模型组、芪白平肺胶囊组,采用烟熏联合低氧法复制COPD痰瘀阻肺证大鼠模型,芪白平肺胶囊组给予1 g/kg芪白平肺胶囊灌胃,正常组和模型组给予等体积生理盐水灌胃。HE染色法观察肺组织形态学特征,ELISA法检测大鼠血清中VEGF水平,荧光定量PCR及Western blot法检测肺血管中Bax和Bcl-2的mRNA和蛋白表达。结果:与正常组相比,模型组肺血管中Bax的mRNA及蛋白表达降低(P<0.01),Bcl-2的mRNA及蛋白表达升高(P<0.01),血清中VEGF表达水平升高(P<0.01);与模型组比,芪白平肺胶囊组Bax的mRNA及蛋白表达升高(P<0.01),Bcl-2的mRNA及蛋白表达水平降低(P<0.01),VEGF表达水平降低(P<0.01)。结论:芪白平肺胶囊可能通过提高COPD痰瘀阻肺证大鼠肺血管中Bax的mRNA及蛋白水平、降低Bcl-2的mRNA及蛋白和血清VEGF水平,调整肺血管的凋亡/增殖失衡状态,从而促进肺血管内皮细胞凋亡,改善COPD肺血管重构。 Objective:To study the relationship between the proliferation/apoptosis related molecule Bax/Bcl-2 and the improvement of pulmonary vascular thickening in rats with chronic obstructive pulmonary diseases(COPD)of phlegm-blood stasis obstructing Lung by Qibai Pingfei Capsule.Methods:SD rats were randomly divided into the normal group,the model group and the Qibai Pingfei Capsule group.COPD model of phlegm-blood stasis obstructing Lung was prepared by fumigating combined with hypoxia method.Qibai Pingfei Capsule group was given Qibai Pingfei Capsule(1g/kg)by gavage,whereas the normal group and the model group were given equal volume of normal saline by gavage.HE staining was used to observe the morphological features of lung tissues.The protein and mRNA expressions of Bax and Bcl-2 in pulmonary vessels were detected by fluorescence quantitative PCR and Western blot,and the serum level of VEGF was detected by ELISA.Results:Compared with those in the normal group,the protein and mRNA expressions of Bax in pulmonary vessels were decreased(P<0.01),the protein and mRNA expression levels of Bcl-2 were increased(P<0.01),and the serum level of VEGF was increased in the model group(P<0.01).Compared with those in the model group,the protein and mRNA expressions of Bax in pulmonary vessels were increased(P<0.01),the protein and mRNA expression levels of Bcl-2 were decreased(P<0.01),and the serum level of VEGF was decreased in the Qibai Pingfei Capsule group(P<0.01).Conclusion:Qibai Pingfei Capsule may regulate the imbalance of apoptosis/proliferation of pulmonary vessels by increasing mRNA and protein levels of Bax and decreasing the mRNA and protein levels of Bcl-2 in pulmonary vessels,as well as reducing the serum level of VEGF in rats with COPD of phlegm-blood stasis obstructing Lung,thereby promoting the apoptosis of pulmonary vascular endothelial cells and improving pulmonary vascular remodeling.
