色季拉山不同海拔高寒草甸土壤线虫群落研究

摘要 为研究不同海拔高寒草甸土壤线虫群落特点,以藏东南色季拉山高寒草甸为研究区,利用高通量测序技术,分析3个不同海拔4600 m(e1)、4500 m(e2)、4400 m(e3)的土壤线虫群落特点。结果表明:在纲水平,Enoplea随海拔升高占比逐渐减少,Chromadorea则在e1占比最多,高达80%。在目水平,e2的线虫目总数多于其他两个海拔,Dorylaimida随海拔升高占比逐渐减少,Rhabditida则随海拔升高占比逐渐增大。同时,e2具有独特的α多样性指数特征,其丰富度与Shannon指数相对较高,但Simpson指数相对较低。利用NMDS分析线虫群落的相似性,发现在纲水平和属水平,e2与e3下的线虫群落较为相似,但e1与这二者的差别较大。由此可见,在不同海拔高寒草甸之间,可能存在海拔分布的差异性,特别对某类线虫如纲水平的Enoplea、目水平的Dorylaimida和Rhabditida等,或许具有海拔响应特殊性。本研究对高寒地带线虫的地理分布格局有了初步探索,为人们认识青藏高原土壤生物提供一定参考。 To study the characteristics of soil nematode communities in alpine meadows at different elevation,the soil nematode communities at three different elevations of 4600 m(e1),4500 m(e2)and 4400 m(e3)were analyzed using high-throughput sequencing technique in the alpine meadows of the Sejila Mountain in southeastern Tibet.The results showed that at the phylum level,the percentage of Enoplea decreased gradually with increasing elevation,while Chromadorea had the highest percentage at e1,up to 80%.At the order level,the total number of nematode order in e2 was more than the other two elevations,and the proportion of Dorylaimida decreased gradually with elevation,while the proportion of Rhabditida increased gradually with elevation.Meanwhile,e2 had a uniqueα-diversity index characteristic,with relatively high richness and Shannon index,but with relatively low Simpson index.The similarity of nematode communities was analyzed using NMDS,and it was found that nematode communities under e2and e3 were more similar at the phylum level and genus level,but e1 was more different from these two.It can be concluded that there may be differences in elevation distribution between alpine meadows at different elevations,especially for certain types of nematodes such as Enoplea at the phylum level and Dorylaimida and Rhabditida at the order level,and perhaps with elevation response specificity.This study provides a preliminary exploration of the geographic distribution pattern of nematodes in alpine zones and provides a definite reference for the understanding of soil organisms on the Tibetan Plateau.
