摘要
目的建立妇安消疹洗液的高效液相色谱(HPLC)指纹图谱。方法采用HPLC法,色谱柱为OMNIBond HPLC Orca C_(18)柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸溶液(含0.1%庚烷磺酸钠),梯度洗脱,流速为1.0 mL/min,检测波长为225 nm或330 nm,柱温为30℃,进样量为10μL。以落新妇苷为参照,测定10批样品的HPLC图谱,采用“中药色谱指纹图谱相似度评价系统(2012.130723版)”评价相似度,确定共有峰及成分。结果10批样品的HPLC图谱有20个共有峰,相似度均大于0.90;确定的5个已知成分为绿原酸、苦参碱、氧化苦参碱、落新妇苷、蒙花苷。结论所建立的指纹图谱可供妇安消疹洗液的真伪鉴别和质量评价参考。
Objective To establish a high-performance liquid chromatagraphy(HPLC)fingerprint of Fu'an Xiaozhen Lotion.Methods The chromatographic column was OMNIBond HPLC Orca C_(18) column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-0.1%phosphoric acid solution(contain 0.1%sodium heptanesulfonate),gradient elution.The flow rate was 1.0 mL/min,the wavelength was 225 nm or 330 nm,the column tempreture was 30℃and the injection volume was 10μL.Astilbin was used as a reference to determine HPLC chromatograms of ten batches of Fu'an Xiaozhen Lotion.The Similarity Evaluation System of Chromatographic Fingerprints of Traditional Chinese Medicine(Version 2012.130723)was used for the similarity evaluation to identify common peaks and components.Results A total of 20 common peaks of the HPLC fingerprint of ten batches of Fuan Xiaozhen Lotion were determined.The similarity was more than 0.90 of each batch.Chlorogenic acid,matrine,oxymatrine,astilbin and buddleoside were determined.Conclusion The established HPLC fingerprint can provide a reference for authenticity identification and quality evaluation of Fu'an Xiaozhen Lotion.
