摘要
目的探讨肺炎支原体肺炎(MPP)病儿血清长链非编码RNAMALAT1(LncRNAMALAT1)、微小RNA-125b(miR-125b)的表达水平及其临床意义。方法选择2016年4月至2018年6月许昌市人民医院40例MMP病儿(MPP组),选择同期该院体检的健康儿童40例(对照组),采用实时荧光定量逆转录聚合酶链反应(qRT-PCR)检测血清LncRANMALAT1、miR-125b的表达水平,酶联免疫吸附测定(ELISA)检测血清白细胞介素-6(IL-6)、肿瘤坏死因子α(TNF-α)水平,比浊法检测血浆C反应蛋白(CRP)水平,比较MPP组同对照组LncRANMALAT1、miR-125b、CRP、IL-6、TNF-α水平差异,并观察MPP病儿不同病程LncRANMALAT1、miR-125b、CRP、IL-6、TNF-α水平差异,分析MPP病儿LncRANMALAT1、miR-125b的表达量与CRP、IL-6、TNF-α的相关性。结果MPP组LncRANMALAT1的表达水平(2.13±0.16)高于对照组(1.00±0.10)(P<0.05),miR-125b的表达水平(0.52±0.11)低于对照组(1.01±0.13)(P<0.05),CRP、IL-6、TNF-α水平高于对照组(P<0.05);重症MPP病儿血清LncRAN MALAT1的表达水平高于轻症病儿(P<0.05),miR-125b的表达水平低于轻症病儿(P<0.05),Pearson相关分析显示,MPP病儿LncRANMALAT1表达量与CRP、IL-6、TNF-α呈正相关(P<0.05),miR-125b的表达量与CRP、IL-6、TNF-α呈负相关(P<0.05)。结论MPP病儿血清LncRNAMALAT1的表达水平明显升高,miR-125b的表达水平明显降低,并且与病情相关,对其进行检测能够为病情评估提供参考。
Objective To investigate the expressions and clinical significance of long-chain non coding RNA malat1(LncRNA MALAT1)and microRNA-125b(miR-125b)in serum on children with Mycoplasma pneumoniae pneumonia(MPP).Methods Forty children with MMP(MPP group)were selected from Xuchang People's Hospital from April 2016 to June 2018,and 40 healthy children(control group)taking physical examination in the same hospital during the same period were selected.The expression levels of serum LncRAN MALAT1 and miR-125b were detected by real-time,fluorescence-based quantitative reverse transcription polymerase chain reaction(qRT-PCR).The levels of serum interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)were detected by enzyme linked im⁃munosorbent assay(ELISA).Turbidimetry was used to detect the levels of C reactive protein(CRP).The difference in the levels of Ln⁃cRAN MALAT1,miR-125b,CRP,IL-6,TNF-αbetween MPP group and control group was compared,and the difference in the levels of LncRAN MALAT1,miR-125b,CRP,IL-6,TNF-αamong MPP children with different courses.The correlation between the expressions of LncRAN MALAT1,miR-125b and those of CRP,IL-6 and TNF-αin MPP children was analyzed.Results The expression level of LncRAN MALAT1 in the MPP group was higher than that in the control group[(2.13±0.16)vs.(1.00±0.10),P<0.05],the expression lev⁃el of miR-125b was lower than that in the control group[(0.52±0.11)vs.(1.01±0.13),P<0.05],and the levels of CRP,IL-6 and TNF-αwere higher than those in the control group(P<0.05).The expression level of serum LncRAN MALAT1 in severe MPP children was higher than that in mild MPP children(P<0.05),and the expression level of miR-125b in severe MPP children was lower than that in mild MPP children(P<0.05).Pearson correlation analysis results showed that LncRAN MALAT1 expression in children with MPP was positively correlated with the expressions of CRP,IL-6,and TNF-α(P<0.05),and miR-125b expression was negatively correlated with the expressions of CRP,IL-6,TNF-α(P<0.05).Conclusions The expression level of LncRAN MALAT1 was significantly increasing and that of miR-125b was significantly decreasing in MPP children,which was related to the condition of the disease.Detection of their expressions could provide reference for disease assessment.
