摘要
目的探讨不同浓度益气化痰活血法代表方剂调脂通脉颗粒对血脂异常痰浊阻遏证小鼠动脉粥样硬化相关炎性因子白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)表达的影响。方法选取雄性ApoE基因敲除小鼠24只,均采用高脂饲料喂养方法构建血脂异常痰浊阻遏证的病-证模型。将造模成功小鼠随机分为模型组、调脂通脉颗粒低剂量组、调脂通脉颗粒中剂量组、调脂通脉颗粒高剂量组,每组6只。除模型组外,调脂通脉颗粒低、中、高剂量组分别给予2 g/(kg·d)、4 g/(kg·d)、8 g/(kg·d)的调脂通脉颗粒灌胃4周。灌胃结束后,采用ELISA法检测各组小鼠血清IL-6、TNF-α水平。结果ApoE基因敲除小鼠均无脱失。模型组、调脂通脉颗粒低剂量组、调脂通脉颗粒中剂量组、调脂通脉颗粒高剂量组小鼠血清IL-6和TNF-α水平均依次下降,差异均有统计学意义(P均<0.05);调脂通脉颗粒高剂量组血清IL-6水平均明显低于模型组、调脂通脉颗粒低剂量组、调脂通脉颗粒中剂量组(P均<0.05),血清TNF-α水平明显低于模型组、调脂通脉颗粒低剂量组(P均<0.05);调脂通脉颗粒低、中剂量组与模型组比较,调脂通脉颗粒低剂量组与调脂通脉颗粒中剂量组比较,血清IL-6和TNF-α水平差异均无统计学意义(P均>0.05)。结论调脂通脉颗粒能降低血脂异常痰浊阻遏证小鼠血清IL-6、TNF-α水平,其中高剂量调脂通脉颗粒可更明显下调IL-6水平,而TNF-α水平的降低对调脂通脉颗浓度的依赖程度不大。调脂通脉颗粒可能通过抑制动脉粥样硬化相关炎性因子的表达,从而减轻血脂异常痰浊阻遏证下血管内皮的炎症反应,延缓病变进展。
Objective It is to investigate the effect of Tiaozhitongmai granule Tiaozhi Tongmai granule,a representative decoction of therapy for invigorating qi,resolving phlegm and activating blood circulation at different concentrations on the expression of atherosclerotic inflammatory factors interleukin-6(IL-6),tumor necrosis factor-α(TNF-α)in dyslipidemia mice with syndrome of phlegm-turbidity stagnation.Methods Twenty-four male ApoE knockout mice were selected and fed with high-fat feed to establish the disease-syndrome models of dyslipidemia with syndrome of phlegm-turbidity stagnation.The successful model mice were randomly divided into model group,Tiaozhi Tongmai granule low-dose group,Tiaozhi Tongmai granule medium-dose group,and Tiaozhi Tongmai granule high-dose group,with 6 mice in each group.Except for the model group,Tiaozhi Tongmai granule low-dose,middle-dose and high-dose groups were respectively treated with 2 g/(kg·d),4 g/(kg·d),and 8 g/(kg·d)Tiaozhi Tongmai granules by gavage for 4 weeks.At the end of treatment,the levels of serum IL-6 and TNF-αof mice in each group were detected by ELISA.Results ApoE knockout mice were not lost.The serum levels of IL-6 and TNF-αin the model group,Tiaozhi Tongmai granule low-dose group,Tiaozhi Tongmai granule medium-dose group,and Tiaozhi Tongmai granules high-dose group decreased sequentially,and the differences were statistically significant(all P<0.05);the level of serum IL-6 in the high-dose group were significantly lower than those in the model group,low-dose group,and medium-dose group(all P<0.05),the level of TNF-αwas significantly lower than that in the model group and the low-dose group(all P<0.05).There were no significant differences in serum IL-6 and TNF-αlevels between the low-dose and medium-dose groups and the model group,and between the low-dose group and the medium-dose group(all P>0.05).Conclusion Tiaozhi Tongmai granules can reduce the levels of serum IL-6 and TNF-αin mice with dyslipidemia and syndrome of phlegm-turbidity stagnation,and high-dose Tiaozhi Tongmai granules can more significantly down-regulate the levels of IL-6,while The decrease of the level of TNF-αhad little dependence on the concentration of Tiaozhi Tongmai granules.Tiaozhi Tongmai granules may reduce the inflammatory response of the vascular endothelium under the dyslipidemia with syndrome of phlegm-turbidity stagnation and delay the progression of the disease by inhibiting the expression of atherosclerosis-related inflammatory factors.
