摘要
目的:研究百会、神庭电针干预脑卒中大鼠的作用以及可能机制。方法:40只脑卒中大鼠随机分为脑卒中组、电针组、L-NAME组、电针+L-NAME组,各10只,并随机选取10只设为假手术组。电针组采用百会、神庭电针干预;L-NAME组建模后1 h、12 h和36 h行L-NAME腹腔注射,每次5 mg/kg;电针+L-NAME组采用百会、神庭电针联合L-NAME干预,操作同电针组与L-NAME组;假手术组与脑卒中组实施非穴刺激。末次干预后4 h Morris水迷宫实验检测认知功能;激光多普勒血流仪检测缺血局部脑血流量;检测脑组织含水量;免疫组化染色观察缺血周边区域新生血管密度;Western blot检测脑组织内皮型一氧化氮合酶(eNOS)、p-eNOS及一氧化氮(NO)蛋白相对表达量。结果:L-NAME组、脑卒中组、电针+L-NAME组、电针组和假手术组逃避潜伏期依次缩短,目标象限停留时间依次延长,穿越原平台次数依次增加,脑血流速度依次加快,脑组织含水量依次减少(P<0.05);脑卒中组神经元数量减少、间隙变大、细胞核固缩、胞浆深染,L-NAME组更为严重,电针组、电针+L-NAME组干预后神经元损伤程度减轻,电针组较电针+L-NAME组改善效果更为明显。假手术组、L-NAME组、脑卒中组、电针+L-NAME组和电针组新生血管密度依次增加(P<0.05),NO蛋白相对表达量、p-eNOS/eNOS依次升高(P<0.05)。结论:百会、神庭电针干预可改善脑卒中大鼠认知功能,加快脑血流速度,缓解脑水肿,减轻海马区病理变化,促进血管新生,推测其作用机制与上调eNOS、NO表达有关。
Objective:To study the therapeutic effect and possible mechanism of electro-needling Baihui(DU20)and Shenting(DU24)in the treatment of stroke rats.Methods:40 stroke rats were randomly divided into the stroke group,the electro-acupuncture group,the L-NAME group,the electro-acupuncture plus L-NAME group,with 10 rats in each group,and another 10 rats were assigned into the sham-operation group.The electro-acupuncture group was treated with electro-needling DU20 and DU24.The L-NAME group was treated with intraperitoneal injection of L-NAME at 1 h,12 h,36 h after modeling,5 mg/kg each time.The electro-acupuncture plus L-NAME group was treated with electro-needling DU20 and DU24 combined with L-NAME injection.The sham-operation group and the stroke group were treated with non-point stimulation.4 h after the last intervention,Morris water maze test was used to measure cognitive function.Laser Doppler blood flow meter was used to measure cerebral blood flow in the ischemic area.Brain tissue water content was measured.The density of angiogenesis was observed by immunocytochemistry.The relative protein expression levels of eNOS,p-eNOS and NO in brain tissue were detected by Western blot.Results:The escape latency was shortened,whereas the staying time in the target quadrant was lengthened,the number of crossing the original platform and the cerebral blood flow speed were increased,and the water content of the brain tissue was decreased in sequence of the L-NAME group,the stroke group,the electro-acupuncture plus L-NAME group,the electro-acupuncture group and the sham operation group(P<0.05).The number of neurons was decreased,the gaps became larger,the nucleus were pyknotic and the cytoplasm were deeply stained in the stroke group,which were more serious in the L-NAME group.After the intervention,the neuron damages were reduced in the electro-acupuncture group and the electro-acupuncture plus L-NAME group,and the improvements were more significant in the electro-acupuncture plus L-NAME group.The density of angiogenesis was increased(P<0.05),and the relative expressions of NO protein and p-eNOS/eNOS were also increased in sequence of the sham operation group,the L-NAME group,the stroke group,the electro-acupuncture plus L-NAME group and the electro-acupuncture group(P<0.05).Conclusion:Electro-needling DU20 and DU24 can improve the cognitive function,accelerate cerebral blood flow,relieve cerebral edema,reduce pathological changes in hippocampus and promote angiogenesis in stroke rats.It is speculated that the action mechanism is related to the up-regulation of eNOS and NO expressions.
