甲状腺功能亢进患者外周血miRNA-21和miRNA-192的表达 被引量:4

Study on the correlation between the expression of miRNA-21 and miRNA-192 in peripheral blood and patients with hyperthyroidism

摘要 目的探讨甲状腺功能亢进(甲亢)患者外周血微小RNA(miRNA)-21和miRNA-192的表达。方法选择甲亢患者97例(甲亢组)作为研究对象;另选择健康体检者80例作为对照组。采用放射免疫分析法测定甲状腺功能指标[游离三碘甲状腺原氨酸(FT3)、总三碘甲状腺原氨酸(TT3)、游离甲状腺素(FT4)、总甲状腺素(TT4)和促甲状腺素(TSH)]水平;采用qRT-PCR法测定外周血miRNA-21和miRNA-192表达。结果甲亢组血清miRNA-21相对表达量高于对照组,而血清miRNA-192相对表达量低于对照组(P<0.05)。甲亢组FT3、TT3、FT4和TT4水平高于对照组,而TSH水平低于对照组(P<0.05)。治疗后血清miRNA-21相对表达量低于治疗前,而血清miRNA-192相对表达量高于治疗前(P<0.05)。miRNA-21对甲亢诊断敏感度和特异度分别为75.44%和70.00%;miRNA-192对甲亢诊断敏感度和特异度分别为80.00%和72.34%。miRNA-21与FT3、TT3、FT4和TT4呈线性正相关,与TSH呈线性负相关;miRNA-192与FT3、TT3、FT4和TT4呈线性负相关,与TSH呈线性正相关。结论甲亢患者中miRNA-21高表达,而miRNA-192低表达,且与甲状腺功能密切相关,可作为诊断和治疗辅助性指标。 Objective To investigate the correlation between the expression of microRNA(miRNA)-21 and miRNA-192 in peripheral blood and patients with hyperthyroidism.Methods 97 patients with hyperthyroidism were selected as the research objects;and 80 patients with healthy physical examination were selected as the control group.Radioimmunoassay was used to determine the level of thyroid function indexes,including free triiodothyronine(FT3),total triiodothyronine(TT3),free thyroxine(FT4),Total Thyroxine(TT4)and thyrostimulating hormone(TSH).RT-PCR method was used to measure the expression of miRNA-21 and miRNA-192 in peripheral blood.Results The relative expression of miRNA-21 in the hyperthyroidism group was significantly higher than control group,while the relative expression of miRNA-192 was significantly lower(P<0.05).The levels of FT3,TT3,FT4 and TT4 in the hyperthyroidism group were significantly higher than those in the control group,while the TSH level was significantly lower than that in the control group(P<0.05).The relative expression of serum miRNA-21 after treatment was significantly lower than that before treatment,while the relative expression of serum miRNA-192 was significantly higher(P<0.05).The sensitivity and specificity of miRNA-21 for the diagnosis of hyperthyroidism were 75.44%and 70.00%,respectively;and the sensitivity and specificity of miRNA-192 for the diagnosis of hyperthyroidism were 80.00%and 72.34%,respectively.miRNA-21 was linearly positively correlated with FT3,TT3,FT4,and TT4,and linearly negatively correlated with TSH;miRNA-192 was linearly negatively correlated with FT3,TT3,FT4,and TT4,and linearly positively correlated with TSH.Conclusion The expression of miRNA-21 is high in patients with hyperthyroidism,while the expression of miRNA-192 is low.It is closely related to thyroid function and can be used as an auxiliary indicator for diagnosis and treatment.
