摘要
目的探讨神经丝蛋白(Neurofilament protein,NFP)及环氧化酶2(Cyclooxygenase-2,COX-2)在子宫内膜异位症(Endometriosis,EMT)及其两种中医证候(气滞血瘀证、肾虚血瘀证)中的表达,以及二者在EMT子宫内膜中表达的相关性。方法取气滞血瘀证、肾虚血瘀证各6例EMT患者的在位和异位子宫内膜及6例正常妇女的在位子宫内膜,共5组,30例标本。使用免疫组织化学S-p法观察各组标本中NFP、COX-2的表达情况。结果(1)NFP在气滞血瘀证在位内膜组和肾虚血瘀证在位内膜组的表达与对照组比较,有显著性差异(P<0.01);在气滞血瘀证异位内膜组和肾虚血瘀证异位内膜组的表达与对照组比较,差异有统计学意义(P<0.05);两证候组在位内膜组间比较,有显著性差异(P<0.01);两证候组异位内膜组间比较,结果无统计学差异(P>0.05)。(2)COX-2在气滞血瘀证在位内膜组和肾虚血瘀证在位、异位内膜组的表达与对照组相比,均有显著性差异(P<0.01);气滞血瘀证异位内膜组的表达与对照组相比,差异有统计学意义(P<0.05);两证候组在位内膜组间比较及异位内膜组间比较,均无统计学差异(P>0.05)。(3)NFP和COX-2在所有EMT患者在位内膜组的表达相关性检验,无统计学意义(P>0.05);在所有异位内膜组的表达相关性检验,无统计学意义(P>0.05)。结论NFP及COX-2在EMT的发生、发展中起到了一定作用,可能成为EMT的诊断标志物,为其治疗提供了潜在的新靶点。其中NFP在EMT不同证候在位内膜组的表达差异具有显著性,可能作为EMT不同证候的特异性蛋白。NFP和COX-2在EMT患者内膜组织中的表达无相关性。
Objective To discuss and explore the expression of neurofilament protein(NFP)and cyclooxygenase-2(COX-2)within endometriosis(EMT),as well as the two relevant TCM syndromes(respectively the syndrome with qi stagnation and blood stasis,syndrome with kidney deficiency and blood stasis),and their correlation in EMT.Methods The eutopic endometrium and ectopic endometrium of 6 patients with EMT of qi stagnation and blood stasis,kidney deficiency and blood stasis,the eutopic endometrium of 6 normal women were selected,with a total of 5 groups and 30 specimens.Then,the immunohistochemical S-P method was adopted to observe the expression of NFP and COX-2 in the sample.Results The expressions of NFP in eutopic endometrium of syndrome of qi stagnation and blood stasis group and in eutopic endometrium of syndrome of kidney deficiency and blood stasis group were significantly different from that in the control group(P<0.01);Both the difference between the expression in ectopic endometrium of syndrome of qi stagnation and blood stasis group and that in the control group and the difference between the expression in ectopic endometrium of syndrome of kidney deficiency and blood stasis group and that in the control group were statistically significant(P<0.05);There was a significant difference between the eutopic endometrium group of the two syndrome groups(P<0.01);There was no statistical difference between the ectopic endometrium group of the two symptom groups(P>0.05).The expressions of COX-2 in eutopic endometrium of syndrome of qi stagnation and blood stasis group and in eutopic and ectopic endometrium of syndrome of kidney deficiency and blood stasis group were all significantly different from that in the control group(P<0.01);Differences between the expressions in ectopic endometrium of syndrome of qi stagnation and blood stasis group and that in the control group were statistically significant(P<0.05);There was no statistical difference both between the eutopic endometrium group and between the ectopic endometrium group of the two symptom groups(P>0.05).There was no statistical significance in the correlation test of the expressions of NFP and COX-2 in all eutopic endometrium of patients with EMT groups(P>0.05);There was no statistical significance in the correlation test of the expressions in all ectopic endometrium groups(P>0.05).Conclusion COX-2 and NFP may are effective in the occurrence and development process of EMT.They may become the diagnostic markers for EMT and provide the treatment of EMT with new target spots.Among them,there are significant difference for the expression of NFP in different syndromes of EMT.In this regard,it may be regarded as the specific protein for differentiation of different syndromes.There is no correlation between the expression of NFP and COX-2 in intima of patients with EMT.
