双向电泳分离水稻叶片蛋白实验教学的探索

摘要 采用双向电泳法分离水稻叶片蛋白,比较和分析了考马斯亮蓝染色方法和硝酸银染色方法对扫描得到的双向电泳图谱的影响。通过改进水稻叶片蛋白的双向电泳分离方法,可以得到更清晰的双向电泳图谱。结果表明,改进后的硝酸银染色方法灵敏度更高,分离蛋白点更多且清晰。由此可知,改进的实验方法可以得到更为灵敏、清晰的双向电泳图谱,能更好地适应和促进双向电泳分离水稻叶片蛋白的实验教学工作。 To improve the method of two-dimensional electrophoresis separation of rice leaf protein and obtain a clearer two-dimensional electrophoresis map,rice leaf protein was separated by two-dimensional electrophoresis.The effect of two dyeing methods(Coomassie brilliant blue staining and silver nitrate staining)on the two-dimensional electrophoresis(2-DE)was compared and analyzed.The results show that the improved staining method(silver nitrate staining method)has higher sensitivity and more protein separation points.The improved experimental method can obtain a more sensitive and clear two-dimensional electrophoresis map,which can better adapt to and promote the experiment teaching of two-dimensional electrophoresis separation of rice leaf protein.
