摘要
目的:优选五味祛脂方提取工艺。方法:采用Box-Behnken响应面法,以得膏率和总黄酮含量为指标,以溶剂量、提取时间、提取次数为考察因素,优选五味祛脂方提取工艺。结果:提取工艺为溶剂量10倍,提取60min,提取3次。得膏率平均值为27.35±1.19 mg/g,RSD为4.36%,总黄酮提取量为54.32±2.24 mg/g,RSD为4.20%,与理论值偏差为-1.32%,接近Box-Behnken响应面试验中的预测值。结论:响应面回归方程拟合度良好,适合用于五味祛脂方提取工艺的回归分析和参数优化。
Objective To Optimization of extraction process of Wuwei Quzhi Prescription(WQP)by Box-Behnken response surface methodology based on CRITIC weighted evaluation.Methods The effects solvent dosage,extraction time and extraction times on the optimization of extraction process of WQP were investigated by multiple indicators comprehensive scoring and Box-Behnken response surface methodology.the content of extraction rate and total flavonoids are the evaluation indexes of extraction process,and the method of analytic hierarchy process was used to calculate the weight coefficient.Results The optimum extraction process was as follow:using 10 times the amount for three times extraction for 60 min.The average extraction rate is 27.35±1.19 mg/g,RSD=4.36%,the total flavonoid extraction is 54.32±2.24 mg/g,RSD=4.20%,which is close to the maximum value in the Box-Behnken response surface methodology.Conclusion The optimum extraction process is simple and feasible,and the extraction efficiency of components is high,which can provide reference for the extraction process of WQP.
