摘要
为研究蒙药古日古木、古日古木-13对Con A致小鼠自身免疫性肝炎的保护作用及对IFN-γ/STAT1/VCAM-1通路的影响。将BALB/c小鼠随机分为空白对照组、模型组、阳性对照组、古日古木低、中、高剂量组、古日古木-13低、中、高剂量组,连续灌胃7 d,于末次给药1 h后,尾静脉注射Con A 20 mg/kg致免疫性肝损伤。酶联免疫吸附测定法测定小鼠血清中肿瘤坏死因子-α(TNF-α)和干扰素-γ(IFN-γ)水平;流式细胞术和TUNEL法测定肝组织细胞凋亡程度;蛋白质印迹检测肝组织STAT1、p-STAT1、VCAM-1表达情况。结果表明:古日古木、古日古木-13可降低Con A所致小鼠免疫性肝损伤小鼠血清中TNF-α、IFN-γ的含量(P<0.05);能够明显抑制Con A引起的肝细胞凋亡;可降低STAT1、p-STAT1、VCAM-1蛋白表达量(P<0.05)。可见古日古木和古日古木-13对Con A所诱导的自身免疫性肝炎具有保护作用,其机制可能与INF-γ/STAT1/VCAM-1信号通路相关。
In order to study the effect of Mongolian medicine Gurigumu and its compound Gurigumu-13 on concanavilin A(Con A)induced autoimmune hepatitis in mice and its influence on INF-γ/STAT1/VCAM-1 pathway.The BALB/c mice were randomly divided into control group,model group,positive group,Mongolian medicine Gurigumu low,middle,high dose groups and Mongolian medicine Gurigumu compound low,middle,high dose groups.After continuous intragastric administration for 7 days,1 hour after the last administration,Con A was used to establish acute liver injury.ELISA was used to detect the contents of TNF-αand IFN-γ,the degree of hepatic cells apoptosis was detected by TUNEL and Flow.Western blot was applied to detect the expression of STAT1,p-STAT1 and VCAM-1.The results show that,Mongolian medicine Gurigumu and its compound Gurigumu-13 can effectively reduce the contents of TNF-αand IFN-γin the blood serum(P<0.05).Inhibits the liver cell apoptosis induced by Con A(P<0.05);decreases the protein expression of STAT1,p-STAT1 and VCAM1(P<0.05).It is concluded that Mongolian medicine Gurigumu and its compound Gurigumu-13 has the function of protectiving Con A-induced acute autoimmune hepatitis,and its mechanism may be related to INF-γ/STAT1/VCAM-1 signaling pathway.
