蓼耳胃肠宁颗粒对模型鼠急性胃炎和小肠蠕动的影响 被引量:3

Effect of Liaoer Weichangning granules on acute gastritis and small intestine peristalsis in model rats

摘要 为了探究蓼耳胃肠宁颗粒对急性胃炎胃损伤的抑制作用和对小肠蠕动功能的调节作用,试验将大鼠分为空白组、模型组和蓼耳胃肠宁高、中、低剂量组及苋菜黄连素组,除空白组外,其他各组先灌胃给予蓼耳胃肠宁或苋菜黄连素药液,再采用95%乙醇溶液灌胃法建立大鼠急性胃炎胃黏膜损伤模型,进行胃组织表面损伤指数和损伤抑制率的测定;利用试剂盒检测胃组织中肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)和白细胞介素8(IL-8)的表达量。将小鼠同样分为6组,建立小鼠大黄型脾虚模型后,灌胃给予蓼耳胃肠宁或苋菜黄连素药液治疗,并于末次给药1 h后对小鼠灌胃给予5%碳末混悬液,检测小鼠小肠内容物推进率。结果表明:与空白组胃组织比较,模型组出现大量条索状出血或出血点。与模型组比较,蓼耳胃肠宁中、高剂量组与苋菜黄连素组胃组织表面出血情况明显减轻;蓼耳胃肠宁中、高剂量组胃黏膜损伤指数显著降低(P<0.05),且损伤抑制率呈一定的剂量依赖关系;蓼耳胃肠宁中、高剂量组的IL-8和IL-6表达量显著降低(P<0.05),蓼耳胃肠宁低、中、高剂量组的TNF-α表达量均显著降低(P<0.05);蓼耳胃肠宁低、中、高剂量组小肠内容物推进率均显著降低(P<0.05)。说明蓼耳胃肠宁颗粒可减轻胃黏膜损伤,抑制胃组织内炎症因子表达,改善小肠蠕动亢进现象,具有抗急性胃炎和调节小肠蠕动功能的作用。 In order to explore the inhibitory effect of Liaoer Weichangning granules on acute gastritis gastric injury and the regulating effect of small intestine peristalsis function, The rats were divided into blank group, model group, Liaoer Weichangning granules high-dose group, medium-dose group, low-dose group and Xiancai Huangliansu group. Except blank group, other groups were given Liaoer Weichangning or Xiancai Huangliansu solution by intragastric administration, a rat model of acute gastric mucosal injury was established by gavage with 95% ethanol solution, and the surface damage index and damage inhibition rate of gastric tissue were measured. The kits were used to detect the expression of tumor necrosis factor α(TNF-α),interleukin 6(IL-6) and interleukin 8(IL-8) in gastric tissue. The mice are equally divided into 6 groups;after the construction of rhubarb-type spleen deficiency mouse model, Liaoer Weichangning and Xiancai Huangliansu liquid was given by gavage for the treatment. After an hour post the last administration, the mice were given 5% carbon powder suspension by gavage, and the advancement rate of the small intestine contents of the mice was detected. The results showed that compared with the gastric tissue of the blank group, the model group had a large number of transverse bleeding or bleeding spots.Compared with the model group, the bleeding on the surface of stomach tissue in Liaoer Weichangning granules medium and high-dose groups and the Xiancai Huangliansu group was obviously reduced;the degree of gastric mucosal injury index in Liaoer Weichangning granules medium-dose and high-dose groups of was significantly reduced(P<0.05),and the results of injury inhibition rate showed a certain dose dependence;the expression levels of IL-8 and IL-6 in Liaoer Weichanging granules medium-dose and high-dose groups of Liaoer Weichangning granules were significantly reduced(P<0.05);the expression levels of TNF-α in Liaoer Weichangning granules low-dose, medium-dose and high-dose groups were significantly reduced(P<0.05);the advancement rates of the small intestine contents in Liaoer Weichanging granules low-dose, medium-dose and high-dose groups were significantly decreased(P<0.05). The results suggested that Liaoer Weichangning granules could reduce gastric mucosal injury, inhibit the expression of inflammatory factors in gastric tissue, and improve the phenomenon of excessive small intestine peristalsis, which could have the effects of anti-acute gastritis and regulating the function of small intestine peristalsis.
