小陷胸汤对高血脂小鼠血脂及免疫功能的影响 被引量:10

The effect of Xiaoxianxiong decoction on blood lipid levels and immune function in hyperlipidemic mice

摘要 为探讨小陷胸汤对高脂血症(hyperlipidemia,HLP)小鼠血脂及免疫功能的影响及其作用机制,准备36只C57BL/6小鼠,通过高脂饲料喂养8周建立HLP模型,随机分为对照组,模型组,小陷胸汤低(0.03 g/mL)、中(0.06 g/mL)、高(0.12 g/mL)剂量组和阳性对照组(2.5 mg/kg洛伐他汀),每组6只。处死小鼠后,称量肝脏、脾脏,计算肝脏指数和脾脏指数;全自动生化分析仪测定小鼠血清总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDL-C)和高密度脂蛋白胆固醇(high density lipoprotein cholesterol,HDL-C)水平;FACS检测CD40^(+)/CD40L^(+)细胞和Th1、Th2百分比;ELISA检测各组小鼠血清CRP表达量。结果显示:与对照组相比,模型组小鼠肝脏指数和脾脏指数升高(均P<0.05);CRP、TC、TG和LDL-C水平上升,HDL-C水平下降(均P<0.05);CD40^(+)/CD40L^(+)细胞、Th1百分比增加,Th2百分比减少(均P<0.05)。与模型组比较,中剂量组和高剂量组小鼠肝脏指数和脾脏指数降低;CRP、TC、TG和LDL-C水平下降,HDL-C水平上升;CD40^(+)/CD40L^(+)细胞、Th1百分比减少,Th2百分比增加(均P<0.05)。由此,小陷胸汤可降低HLP小鼠血脂水平,减轻机体炎症反应水平并提高免疫功能。 In order to investigate the effect of Xiaoxianxiong decoction on lipidemia and immune function of hyperlipidemia(HLP)mice and the underlying mechanism,36 C57 BL/6 mice were fed with high-fat diet for 8 weeks to establish the HLP model.The mice were randomly divided into control,model,Xiaoxianxiong decoction low-dose(0.03 g/mL),medium-dose(0.06 g/mL),high-dose(0.12 g/mL)and positive control(2.5 mg/kg lovastatin)groups,with 6 mice per group.After mice sacrifice,the liver and spleen were weighed,and the liver index and spleen index were calculated.The levels of total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C),and high density lipoprotein cholesterol(HDL-C)in the mice serum were measured by automatic biochemical analyzer.The proportion of CD40^(+)/CD40 L^(+)cell and Th1,Th2 were detected by FACS,and serum CRP expression was detected by ELISA.The results showed that compared with the control group,the liver and spleen index of the model group increased(all P<0.05);The levels of CRP,TC,TG,and LDL-C increased(all P<0.05),and the level of HDL-C decreased(P<0.05);The percentages of CD40^(+)/CD40 L^(+)cell and Th1 increased and the proportion of Th2 decreased(all P<0.05).Compared with the model group,the mice liver and spleen index in the middle-dose and high-dose groups were decreased(all P<0.05);The levels of CRP,TC,TG,and LDL-C were decreased(all P<0.05),while the level of HDL-C was increased(P<0.05);The percentages of CD40^(+)/CD40 L^(+)cell and Th1 were decreased,and theTh2 percentages were increased(all P<0.05).Therefore,Xiaoxianxiong decoction can reduce the blood lipid levels in HLP mice,reduce the degree of inflammation and improve the immune function.
