摘要
目的:探究核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)炎症小体对糖尿病性视网膜病变新生血管形成和血管拟态标志物的影响。方法:选取65例增殖性玻璃体视网膜病变患者,且行睫状体扁平部23G标准三通道玻璃体切割术的者作为研究对象,经内科检查,其中38例患者确诊为2型糖尿病,作为试验组;27例患者为非糖尿病患者,作为对照组。比较两组患者新生血管密度、NLRP3炎症小体表达水平和血管拟态标志物水平。结果:试验组患者新生血管数为+~++级别的患者高于对照组,而级别的患者则显著小于对照组;试验组患者新生血管密度显著小于对照组,差异具有统计学意义(均P<0.01);试验组患者NLRP3和Caspase-1相对表达水平、血管拟态标志物VEGF-A、VE-cadheri、EphA2、MMP-2、MMP-9表达水平显著高于对照组,差异均有统计学意义(均P<0.01)。结论:NLRP3炎症小体显著影响糖尿病性视网膜病变晚期新生血管形成和血管拟态标志物的表达。
Objective:To investigate the effect of NLRP3 inflammasome on the neovascularization and vascular mimetic markers in diabetic retinopathy.Methods:65 patients with proliferative vitreoretinopathy who underwent 23G standard three-channel vitrectomy on the flat part of the ciliary body were selected as the research objects.After medical examination,38 patients were diagnosed with type 2 diabetes as the experimental group,27 patients without diabetes served as control group.The density of new blood vessels,the expression levels of NLRP3 inflammasome and the vascular mimicry markers were compared between the two groups.Results:The number of patients with neovascularization in grade + to ++ in the experimental group was higher than that of the control group,while the number of patients with neovascularization in grade was significantly lower than that of the control group,and the neovascularization density was significantly lower than that of the control group(all P<0.01).The relative expression levels of NLRP3 and Caspase-1,the expression levels of vascular mimicry markers VEGF-A,VE-cadheri,EphA2,MMP-2,and MMP-9 in the experimental group were significantly higher than those in the control group(all P<0.01).Conclusion:NLRP3 inflammasome significantly affects the late neovascularization of diabetic retinopathy and the expression of vascular mimicry markers.
